冻存对经血源性间充质干细胞表面标记的影响

【摘要】 目的比较冻存前后经血源间充质干细胞(MMCs)3种表面标志表达的变化,分析冻存条件对其表达的影响。方法淋巴细胞分离液法分析经血中单个核细胞,经过体外原代和传代培养,筛选具有增殖能力的细胞。观察细胞形态,流式细胞术分析冻存前后分析表面标志(CD105、CD14和CD45)变化。结果经血单个核细胞传代3次后可获得具有间充质干细胞形态特点MMCs;MMCs高表达CD105分子,低表达CD14和CD45;冻存不影响MMCs表型。结论自经血中可获得MMCs,冻存对MMCs表型CD105high、CD14low、CD45low无影响。

【Abstract】 Objective To analysisthe change of 3 cell surface markers on menstrual blood-derived mesenchymal stem cells( MMCs) and study the expression of cell membrane markers on menstrual blood-derived mesenchymal stem cells affacted by cryopreservation. Methods The mononuclear cells were separated from menstrual blood by lymphocyte separation medium. After primary and continuous cell culture,the high proliferating subgroup was obtained from the mononuclear cells. The cells morphology was observed by microscope and the cell surface markers( CD105,CD14 and CD45) were measured by flow cytometry before or after cryopreservation. Results The MMCs liked could be obtained from menstrual blood mononuclear cells after 3 times continuous culturing. It was found that these MMCs expressed high level CD105,but the expression of CD14 or CD45 was weak. The cryopreservation had no influenceon cell phenotype of MMCs. Conclusion The MMCs could be obtained from menstrual blood,and the cell phenotype CD105 high,CD14low,CD45low do not been affacted by cryopreservation.
