完整基因表达模型中的精确概率分布
Exact distributions in the full models of gene expression
【摘要】 基因表达是细胞内部过程的中心环节,其建模与分析一直是人们关注的焦点.转录水平上的两类代表性基因表达模型(即构成式表达模型和爆发式表达模型)已经被广泛研究,特别是m RNA的分布已经解析地给出,但对于同时考虑转录和翻译的完整基因表达模型,如何解析地导出m RNA和蛋白质的分布以及它们的联合分布的问题至今未得到解决.本文应用作者自己发展起来的二项矩方法解决了此数学问题,尤其是给出了基因产物分布及有关统计量的解析表示.此外,本文还讨论了基因表达噪声的可调性.
【Abstract】 Gene expression is the core of intracellular processes, and its modeling and analysis has been being a focus. The two-state model of gene expression at the transcription level has been extensively studied, and m RNA distribution has analytically been derived, but questions of, e.g., how m RNA and protein distributions and the joint distribution of m RNA and protein in the full gene model that considers transcription and translation simultaneously, remain unsolved. This paper thoroughly solve these mathematical questions by using a binomial moment approach. In particular, formula for calculating these distributions and their statistical quantities are analytically derived. In addition, tunability of gene expression noise is discussed.
