限食对肥胖大鼠下丘脑腹内侧核瘦素、胃动素受体的影响

【摘要】 目的探讨限食对谷氨酸钠诱导的肥胖大鼠下丘脑腹内侧核区瘦素和胃动素受体表达的影响。方法新生雄性Wistar大鼠90只:随机分为对照组30只、模型组60只,模型组皮下注射谷氨酸钠建立肥胖模型。8周时把模型组随机平分为限食组和肥胖组,限食组30%能量限制。16周取材,测定肥胖相关指标;取下丘脑行免疫组化,观察下丘脑腹内侧核瘦素、胃动素受体的表达。应用SPSS16.0软件,所获数据采用方差分析和行配对资料t检验。结果肥胖组与对照组Lee’s指数、各部脂肪含量、血清总胆固醇、甘油三酯、瘦素、胃动素的含量比较,P均<0.0005。限食组与对照组Lee’s指数、各部脂肪含量、血清总胆固醇、甘油三酯、瘦素、胃动素的含量比较,P均<0.0005。三组大鼠下丘脑腹内侧核瘦素、胃动素受体阳性细胞表达的比较,P均<0.0005。结论限食可以改善谷氨酸钠诱导的肥胖指标,可能是与下丘脑腹内侧核区瘦素表达减少、胃动素的表达增强有关。

【Abstract】 Objective To investigate the effects of diet restriction on leptin and motilin receptors’ expression in the hypothalamus of the obese rats induced by glutamate. Methods The male newborn Wistar mice: 30 mice in the control group and 60 in the model group, and the obese model accepted subcutaneous injection of glutamate. The model group was randomly divided into the diet restriction group and the obese group at the end of 8 weeks. The diet restriction group got 30 percents of diet restriction. At the end of 16 weeks, we measured the related obese indicators, did hypothalamic immunohistochemistry and observed the expression of leptin and motilin receptor in hypothalamus. Using SPSS16.0 software, the data was tested by variance analysis and paired t test. Results The Lee’s index, total fat content, serum total cholesterol, triglyceride, leptin, and motilin were compared between the obese group and the control group, with P<0.0005. The comparison between the diet restriction group and the control group were with P<0.0005. The expression of leptin and motilin receptors’ positive cells in the hypothalamus among the three groups were compared with P<0.0005. Conclusion The diet restriction can improve the obese index induced by sodium glutamate, which may be related to the decrease of leptin expression and the increase of motilin expression in the ventral region of hypothalamus.
