固定化酵母菌和米曲霉生物转化黄芩的实验研究

【摘要】 目的筛选黄芩生物转化的单一菌株和复合菌株,并优选出固定化复合菌体系及生物转化黄芩素和汉黄芩素的发酵条件。方法利用微生物转化法和高效液相色谱法,筛选出用于固定化的复合菌株体系;通过单因素实验和正交实验确定黄芩转化的最优条件。结果经固定化的酵母菌和米曲霉生物转化黄芩,其黄芩苷和汉黄芩苷的平均转化率分别高达91.62%和90.02%,黄芩素和汉黄芩的平均提取率分别提高了3.46倍和3.04倍。最优条件:pH为5.0,温度为33℃,时间为48 h,接菌量为12g/瓶,菌液浓度为1.5×107/m L,液料比为30∶1 m L/g。结论固定化复合菌株生物转化黄芩的优选转化条件稳定可行,可为其工业化生产提供理论依据。

【Abstract】 Objective To obtain the fungal strains for biotransformation of Scutellaria Radix roots,and to optimize biotransformation process of baicalein and wogonin by immobilized costrains. Methods The immobilized co-strains were screened by biotransformation and high performance liquid chromatography(HPLC). Single factor tests and orthogonal test were employed to determine the optimization of fermentation conditions of the Scutellaria Radix. Results Scutellaria Radix was transformed by immobilized yeast and Aspergillus oryzae,and the average transformation ratio of baicalin and wogonoside reached 91. 62% and 90. 02%,respectively. Hence,the average yield of baicalein and wogonin were about 3. 46-fold and 3. 04-fold higher than those of the raw samples without transformation,respectively. The optimal immobilization conditions were: pH of 5. 0,incubation temperature of 33 ℃,fermentation time of 48 hours,the concentration of bacterium of 1. 5 × 107cells/m L,the wet weight of immobilized costrains of 12 g and ratio of liquid to solid of 30∶ 1 m L/g. Conclusion The optimal conditions which transformation of Scutellaria Radix roots by the immobilized co-strains are stable and feasible,and could provide experimental basis for the industrial production.
