外周血中树突状细胞及相关细胞因子水平与Graves病的关联分析

【摘要】 目的检测Graves病患者在治疗前后外周血中树突状细胞(DCs)不同亚群和相关细胞因子的数量和表达情况,并与健康对照组进行比较,分析DCs细胞亚群和相关细胞因子与Graves病的关联性。方法利用流式细胞仪检测56例Graves病患者和30名健康体检者的外周血树突状细胞亚群数量,其中Graves病患者中30人为治疗前组、26人为治疗后组,同时利用ELISA检测肿瘤坏死因子-α(TNF-α)、干扰素-γ(IFN-γ)、白细胞介素-2(IL-2)及白细胞介素-6(IL-6)等细胞因子的水平。结果测定得出Graves病治疗前组浆细胞样树突状细胞(m DC)和髓样树突状细胞(p DC)值显著高于健康对照组,经治疗后m DC和p DC值明显降低,略高于健康对照组。通过对比3组细胞因子水平发现,Graves病治疗前组患者血清中TNF-α、IFN-γ、IL-6水平均高于其他2组,治疗后组与健康对照组的细胞因子水平相近。结论 Graves病患者血中树突状细胞(DC)明显升高,细胞因子分泌失调,经治疗病情好转后,DC和细胞因子水平逐渐恢复正常,提示DC和细胞因子可能参与Graves病的发病机制,并有望作为分析Graves病治疗效果的一项依据。

【Abstract】 Objective To explore the changes of different DC subsets and the secretion of cytokines level in the peripheral blood of Graves’ disease patients compared to the healthy controls. Then the correlation of them were analyzed. Methods Flow cytometry was applied to assess the frequency of different DC subsets in peripheral blood of 56 Graves disease patients and 30 healthy cases respectively. 56 Graves disease patients were divided into the before treatment group(30 cases) and the after treatment group(26 cases). Moreover, ELISA was performed to detect the secretion of TNF-α,IFN-γ, IL-2 and IL-6 in the peripheral blood. Results The frequency of p DCs and m DCs in the before treatment group was higher than in healthy cases and it was significantly decreased after treatment. The secret of TNF-α,IFN-γ and IL-6 in Graves disease patients’ peripheral blood were higher than that in other groups. Nevertheless, them were found similar levels of the after treatment group and healthy cases. Conclusion Dendritic cells are increased significantly and the cytokines secretion disorder in the blood of Graves patients. However, they can be decreased by treatment, which indicates that DC subsets and the secretion of cytokines level may be involved in the pathogenesis of Graves. They are hopefully used as the basis for analysis of Graves disease treatment effect.
