【摘要】 目的:探讨温阳活血方对慢性心力衰竭大鼠心肌纤维化的影响。方法:利用肾上腹主动脉缩窄法制作慢性心力衰竭大鼠模型。将30只雄性wistar大鼠随机分为假手术组、模型组、温阳活血方组,每组10只。温阳活血方组给予温阳活血方8m L/kg灌胃,其他两组给予同等剂量的0.9%氯化钠注射液灌胃,连续给药4周后观察各组大鼠左心室质量指数(LVMI)和胶原容积积分(CVF)水平,实时荧光定量(RT-PCR)测定各组大鼠左心室心肌结缔组织生长因子(CTGFmRNA)水平。结果:与假手术组比较,模型组LVMI、CVF及左心室心肌CTGFmRNA表达水平升高(P<0.05,P<0.01);与模型组比较,温阳活血方组LVMI、CVF及左心室心肌CTGFmRNA表达水平下降(P<0.05,P<0.01)。结论:温阳活血方能明显改善心肌纤维化,这可能与抑制CTGF过度表达有关。更多还原

【Abstract】 Objective: To investigate the effect of Wenyang Huoxue prescription on myocardial fibrosis in rats with chronic heart failure. Methods: A rat model of chronic heart failure was established by coarctation of the suprarenal abdominal aorta. A total of 30 male Wistar rats were randomly divided into sham-operation group,model group,and Wenyang Huoxue prescription group,with 10 rats in each group. The rats in the Wenyang Huoxue prescription group were given Wenyang Huoxue prescription 8 ml/kg by gavage,and those in the other two groups were given 0. 9% sodium chloride injection at the same dose by gavage. After administration for 4 consecutive weeks,leftventricular mass index( LVMI) and collagen volume fraction( CVF) were observed,and RT-PCR was used to measure the mRNA expression of connective tissue growth factor( CTGF) in the left ventricle. Results: Compared with the sham-operation group,the model group had significant increases in LVMI,CVF,and mRNA expression of CTGF( P < 0. 05,P < 0. 01). Compared with the model group,the Wenyang Huoxue prescription group had significantreductions in LVMI,CVF,and CTGF mRNA expression( P < 0. 05,P < 0. 01). Conclusion: Wenyang Huoxue prescription can significantly improve myocardial fibrosis,possibly by inhibiting the overexpression of CTGF.更多还原
