IL-1β或TNF-α干预的子宫蜕膜细胞对母胎界面Th1/Th2及Th17/Treg平衡的影响

【摘要】 目的:探讨IL-1β或TNF-α干预的子宫蜕膜细胞对母胎界面Th1/Th2及Th17/Treg平衡的影响。方法:子宫蜕膜细胞经体外分离后,加入10–8 mol/L雌二醇(E2)+10–7 mol/L孕激素(P)培养7 d后,分别给予IL-1β或TNF-α干预24 h,收集上清液,制备条件培养液。应用条件培养液刺激子宫蜕膜T细胞72 h后,收集上清液,ELISA法检测上清液IFN-γ,IL-2,IL-17,IL-5,IL-10及TGF-1β的表达。收集子宫蜕膜T细胞,提取RNA,用实时定量PCR检测细胞因子m RNA的表达。通过T细胞分泌的细胞因子水平来评估子宫蜕膜细胞对Th1/Th2及Th17/Treg平衡的影响。结果:IL-1β或TNF-α干预子宫蜕膜细胞24 h后,其条件培养液能够促使子宫蜕膜T细胞Th1细胞因子IFN-γ分泌增加7~8倍,IL-2分泌增加6~7倍,Th17细胞因子IL-17分泌增加15~19倍,Th2细胞因子IL-10和IL-5的分泌无差异,Treg细胞因子TGF-1β的分泌无差异;其条件培养液促使IFN-γ和IL-17 m RNA表达增加,而IL-10及TGF-1βm RNA的表达无差异。结论:IL-1β或TNF-α刺激的子宫蜕膜细胞改变母胎界面Th1/Th2及Th17/Treg细胞的平衡,向Th1及Th17偏移。

【Abstract】 Objective: To investigate the effect of IL-1β or TNF-α-treated human decidual cells on the balance between Th1 and Th2 or Th17 and Treg.Methods: Human decidual cells were isolated and treated with estrogen(E2) and progesterone(P) for 7 days and then with IL-1β or TNF-α for 24 h. The culture medium was collected to prepare conditioned medium. The T cells isolated from human decidual tissue were cultured in presence of conditioned medium for 72 h. ELISA was used to detect concentration of IFN-γ, IL-2, IL-5, IL-17 IL-10, and TGF-1β in supernatants and quantitative real-time PCR was used to detect the m RNA expression of IFN-γ, IL-17, IL-10, and TGF-1β.Results: The levels of IFN-γ, IL-2 and IL-17 were significantly increased when T cells were cultured in the conditioned medium compared with the control group(E2+P), while the levels of IL-5, IL-10, and TGF-1β were not significantly changed. The m RNA expressions of IFN-γ and IL-17 m RNA were significantly increased when T cells were cultured in the conditioned medium compared with the control group(E2+P), while the m RNA expression of IL-10 and TGF-1β was not significantly different.Conclusion: Human decidual cells treated by IL-1β or TNF-α can shift the balance of Th1 and Th2 or Th17 and Treg toward Th1 and Th17 immunity.
