脂氧素A4对缺氧状态下人肝癌细胞增殖的影响

【摘要】 目的探讨缺氧环境下脂氧素A4(lipoxin A4,LXA4)对肝癌细胞增殖的影响。方法采用小鼠肝癌细胞H22接种构建小鼠肝癌皮下移植瘤模型,观察LXA4对肿瘤生长的影响。采用免疫组织化学法检测细胞增殖核抗原Ki-67和缺氧诱导因子-1α(hypoxia-inducible factor-1α,HIF-1α)表达。四甲基偶氮唑盐比色法检测细胞增殖水平。结果在接种肝癌细胞H22的715 d,小鼠皮下可见清晰肿瘤包块生长;在各相应时间点,给予LXA4的小鼠肿瘤体积均明显缩小(P<0.05),同时明显降低了肿瘤组织内部Ki-67和HIF-1α表达。体外培养发现200 nmol/L LXA4可明显抑制缺氧条件下人肝癌细胞HepG2的增殖。结论 LXA4能抑制缺氧条件下肝癌细胞的增殖。

【Abstract】 Objective To explore the effect of lipoxin A4(LXA4) on the proliferation of human hepatocellular carcinoma(HCC) under hypoxic condition.Methods Ectopic HCC mice model was established by injection H22 cells subcutaneously.Immunohistochemistry assay was applied to detect the protein expression of Ki-67 and hypoxia-inducible factor-1α(HIF-1α).HepG2 cells were cultured under 1%O2 to establish a hypoxic condition.3-(4,5-dimethyl-2-thiazolyl)-2,5-diphenyl-2-H-tetrazolium bromide(MTT) assay was used to measure cell proliferation rate.Results In 7 to 15 days after H22 cell transplantation,the subcutaneous tumor in mice could be observed.At all the time points,the tumor volume of LXA4 mice was significantly reduced(P<0.05).At the same time,LXA4 significantly decreased the expression of Ki-67 and HIF-1α inside tumor tissue.In vitro study showed that 200 nmol/L LXA4 obviously inhibited HepG2 proliferation under 1%O2.Conclusion LXA4 could inhibit the HCC growth under hypoxia.
