鸡骨香中的抗癌化学成分研究
Study on the anti-cancer chemical compositions from croton crassifolius geisel
【摘要】 研究了鸡骨香中的抗癌类化学成分。通过硅胶柱色谱及高压液相色谱等方法对鸡骨香进行分离,根据理化性质和光谱数据鉴定化学结构,应用高通量筛选模型评价化合物的抗癌活性。结果表明,从鸡骨香中分离得到7个黄酮类化合物,分别鉴定为千层纸素A(Ⅰ)、汉黄芩素(Ⅱ)、黄芩新素Ⅱ(Ⅲ)、黄芩素(Ⅳ)、黄芩苷(Ⅴ)、白杨素-6-C-α-L-吡喃阿拉伯糖-8-C-β-D-吡喃葡萄糖苷(VI)、白杨素-6-C-β-D-吡喃葡萄糖-8-C-α-L-吡喃阿拉伯糖苷(Ⅶ);其中化合物Ⅱ、Ⅲ、Ⅳ对肿瘤细胞Sw-620、Hela、A549、MDA-MB-231均有显著的抑制作用。7个化合物均为首次从鸡骨香中分离得到,化合物Ⅱ、Ⅲ、Ⅳ显示有体外抗肿瘤活性。
【Abstract】 To study the anti- cancer chemical compositions from croton crassifolius geisel,the compounds were isolated by silica gel column chromatography and HPLC,their structures were identified according to their physical and chemical properties and spectral data,their anti- cancer activity were evaluated using high- throughput screening model.The results show thatseven compounds were isolatedand identified as oroxylin A(I),wogonin(Ⅱ),skallcapflavon Ⅱ(Ⅲ),baicalein(IV),baicalin(V),chrysin- 6- C- α- L- arabinopyranosyl- 8- C- β- D- glucopyranoside(VI),chrysin- 6- C- β- D- glucopyranoside- 8- C- α- L- arabinopyranosyl(Ⅶ).Among them,compoundⅡ,compound Ⅲand compound IV all showed strong inhibitory activity against human cancer Sw- 620,Hela,A549 and MDA- MB- 231.Seven flavonoids were all isolated from croton crassifolius geisel for the first time,and compound Ⅱ、Ⅲ、IV have significant anti- cancer activity.
