外施五氮杂胞苷对菊花生理生化及DNA甲基化和基因差异表达的影响
Effect of External 5-azaC on Physiology and DNA Methylation and Gene-Expression of Chrysanthemum
【摘要】 本研究采用不同浓度的5-氮杂胞嘧啶核苷(5-azacytidine,5-azaC)在体外处理菊花品种"泉乡冲天",研究其表型、DNA甲基化水平及基因表达差异.结果表明,100、200、400mmol/L的5-azaC处理对菊花株高有明显的抑制作用,且200mmol/L处理时花期提前.甲基化敏感扩增多态性分析(MSAP)及高效液相色谱分析(HPLC)结果表明:处理组与对照组相比甲基化都有所降低,降低比率分别为12.40%、19.30%和23.00%.cDNA扩增片段长度多态性分析显示,提前开花的菊花特异性表达片段30条,成功测序的10条中,7条可以在NCBI中找到同源序列,为已知功能基因,其功能主要是参与信号转导、蛋白质代谢和抗病.以上结果表明,采用甲基化抑制剂5-azaC处理菊花可使其基因组甲基化水平降低,导致提前开花,且基因表达存在一定的差异.
【Abstract】 The objectives of this research were to assess the effect of 5-azacytidine(5-azaC)stress on the phenotype as well as genomic DNA methylation levels and gene differentially expression of chrysanthemum seedlings.The experiment results show that,100、200、400 mmol/L of 5-azaC inhibite the plant height of chrysanthemum,and flowering-time of the 200mmol/L is earliest in all of materials.The results of MSAP and HPLC indicate that the genomic of treated materials DNA methylation decreases distinctly,and the ratios of DNA methylation reduction are12.40%,19.30% and 23.00%,respectively.The amplified fragment length polymorphism of cDNA shows that among 30 gene specific expressions of the precocious flowering,there are ten successful expressions,seven of which have homologous series in the National Center of Biotechnology Information.They are known functional gene involving in signal transduction,protein metabolism and disease resistance.Based on these,the effects of 5-azacytidine,a methylation inhibitor,on chrysanthemum lower methylation content resulting in precocious flowering and gene expression differently.
