乙氧氟草醚对泥鳅的毒性研究

【摘要】 为了检测除草剂乙氧氟草醚对水生生物的毒性,以泥鳅(Misgurnus anguillicaudatus Cantor)为试验对象,通过急性毒性试验、微核试验、单细胞凝胶电泳试验,来检测其对泥鳅的毒性效应。结果表明,乙氧氟草醚对泥鳅在24、48、72、96 h的半数致死浓度(LC50)分别为36.66、32.86、28.92、27.39 mg/L,安全浓度(SC)为7.92 mg/L。微核率和核异常率均随着染毒浓度的增加和时间的延长不断升高,呈现明显的时间剂量效应关系。在处理6 d的最高浓度,微核率和核异常率达到最大,分别为6.417‰±0.631‰和65.417‰±5.080‰。单细胞凝胶电泳试验中,选取彗尾DNA含量、尾长、尾矩3个指标作为检测DNA受到损伤的认定标准,结果处理6 d的16.00 mg/L浓度达到峰值,单细胞彗尾DNA含量为43.06%±2.54%,尾长为268.00 pix±8.49 pix,尾矩为89.31±1.82。试验结果为乙氧氟草醚的合理使用以及环境污染监测提供了科学依据。

【Abstract】 In order to estimate the herbicide toxicity of oxyfluorfen to aquatic organisms,regarding the Misgurnus anguillicaudatus Cantor as the experimental object,we adapted the acute test,micronucleus test,single cell gelelectrophoresis to test its toxicity on M.anguillicaudatus.The results showed that,the half lethal concentration(LC50) of oxyfluorfen on the24,48,72,96 h were 36.66,32.86,28.92,27.39 mg/L;respectively;the safe concentration(SC) was 7.92 mg/L.The micronucleus rate and nuclear anomalies in each experiment were increased with exposure time and concentration,which presents obvious time dose effect.The highest micronuclei and nuclear anomalies’ rate reached the maximum which appeared in the highest concentration on 6 d.The results were permill 6.417‰±0.631‰,65.417‰±5.080‰,respectively.In the comet assay,we chose the content of comet tail DNA,tail length and the olive tail moment(these three standards) to check the injure degree of DNA.The results showed that they reached to the peak value on 6 d and 16.00 mg/L.The peak value was43.06% ±2.54%,268.00 pix ±8.49 pix,89.31 ±1.82,respectively.The studies showed scientific references for effective use of oxyfluorfen and environmental pollution monitoring.
