新疆沙枣树叶提取物对糖尿病小鼠血糖的影响

【摘要】 用溶剂提取法制备新疆沙枣树叶的提取物,得到沙枣树叶乙酸乙酯提取物、沙枣树叶正丁醇提取物和沙枣树叶水提取物;用体外α-葡萄糖苷酶抑制模型,对3种沙枣树叶提取物进行α-葡萄糖苷酶抑制活性筛选,并与阳性对照药阿卡波糖进行比较。采用腹腔注射四氧嘧啶构建糖尿病动物模型,造模成功后,将糖尿病小鼠分为模型对照组、二甲双胍处理组、新疆沙枣树叶乙酸乙酯提取物和水提取物处理组,另设正常对照组。连续灌胃给药18 d,每天一次,采血测空腹血糖。结果表明,新疆沙枣树叶的3种提取物对α-葡萄糖苷酶的活性有较好的抑制作用,有2个提取物的抑制率明显高于阿卡波糖,其中新疆沙枣树叶乙酸乙酯提取物活性最强,在浓度为0.075 g/L时,其酶抑制率为76.62%。阿卡波糖及3种提取物对α-葡萄糖苷酶抑制作用的IC50分别为阿卡波糖IC50891.38 mg/L,沙枣树叶乙酸乙酯提取物IC509.28mg/L,沙枣树叶正丁醇提取物IC50103.5 mg/L,沙枣树叶水提取物IC5073.99 mg/L。结果显示,新疆沙枣树叶提取物对四氧嘧啶所致糖尿病小鼠的血糖有降低的作用,和对照组比较有显著差异(P<0.05)。新疆沙枣树叶提取物能显著抑制α-葡萄糖苷酶活性,对糖尿病小鼠具有降糖作用。

【Abstract】 Three kinds of extracts of Xinjiang Elaeagnus angustifolia L. Leaves(EAL) including EAL ethylacetate extraction,EAL normal butyl alcohol extraction and EAL water extraction were prepared by solvent extraction. The inhibitory effects of these extracts to α-glucosidase activity were investigated by in vitro enzymatic method;and akarbose was used as positive reference substance. The diabetic model were induced by peritoneal injection of alloxan. Then the diabetic mice were divided into model control group,metformin treated group,EAL ethylacetate extraction treated groups,EAL water extraction treated groups and normal control group was another one. The drugs were chronically administrated to diabetic mice once a day for18 days. The blood glucose(GLu) were determined. The results showed that, the extracts of EAL showed obviously inhibitory action against α-glucosidase activity in which three extracts have higher effects compared to those of acarbose,among them,the extract of EAL ethylacetate extraction was more effective for enzyme inhibition with a inhibitory rate of 76.62%. The EAL IC50 of the three extracts were:Acarbose IC50891.38 mg/L,EAL ethylacetate extraction IC509.28 mg/L,EAL normal butyl alcohol extraction IC50103.5 mg/L and EAL water extraction IC5073.99 mg/L. The result indicate that EAL extraction has the effect which can lower the level of blood sugar of tetroxide pyrimidine diabetes mice,comparing with control group,the difference is obviously(P<0.05). Xinjiang E. angustifolia L. Leaves extracts had remarkable inhibitory effect to α-glucosidase activity. The EAL can decrease the blood glucose.
