五味子乙素通过上调miR-486-5p的表达对甲状腺癌细胞生物学行为的影响
Effect of Schisandrin B on Cell Biological Behavior of Thyroid Carcinoma by Up-regulating MiR-486-5p Expression
【摘要】 目的研究五味子乙素对miR-486-5p表达及人甲状腺癌B-CPAP细胞增殖和侵袭的影响。方法实时定量PCR检测人甲状腺癌B-CPAP细胞中miR-486-5p的表达量,Western-blot法检测人甲状腺癌B-CPAP细胞中Caspase-3、Bcl-2和Bax的表达。脂质体转染方法构建过表达miR-486-5p的人甲状腺癌B-CPAP细胞,MTT法检测人甲状腺癌B-CPAP细胞的增殖率,流式细胞仪检测人甲状腺癌B-CPAP细胞的凋亡率。结果与空白对照组比较,随着五味子乙素浓度的增加人甲状腺癌B-CPAP细胞增殖率和Bcl-2表达呈不断降低趋势(P<0.05),miR-486-5p、Caspase-3和Bax表达呈不断增加趋势(P<0.05)。转染miR-486-5p模拟物组的细胞增殖率在转染3 h与4 h后低于转染miRNA模拟物组,细胞凋亡率显著高于转染miRNA模拟物组(P<0.05,P<0.01)。结论五味子乙素可能通过上调miR-486-5p表达,诱导人甲状腺癌B-CPAP细胞发生凋亡,miR-486-5p可作为对人甲状腺癌靶向治疗的靶点。
【Abstract】 Objective To investigate effects of Schisandrin B on miR-486-5p expression and proliferation and invasion of human thyroid carcinoma B-CPAP cells. Methods Real-time quantitative polymerase chain reaction( PCR)was used to detect miR-486-5p expression in human thyroid carcinoma B-CPAP cells,and Caspase-3,Bcl-2 and Bax expressions in human thyroid carcinoma B-CPAP cells were detected by western-blot method. Liposome transfection method was used to establish human thyroid carcinoma B-CPAP cells with overexpressing miR-486-5p. Proliferation rate of human thyroid carcinoma B-CPAP cells was detected by MTT assay,and apoptotic rate of human thyroid carcinoma B-CPAP cells was detected by flow cytometry. Results Compared with those in control group,proliferation rate and Bcl-2 expression of human thyroid carcinoma B-CPAP cells were gradually decreased( P < 0. 05),while miR-486-5p,Caspase-3 and Bax expressions were gradually increased with increasing concentration of Schisandrin B( P < 0. 05). Cell proliferation rates of transfecting miR-486-5p mimics at 3 h and 4 h after transfection were significantly lower than those of transfecting miRNA mimics,while apoptotic rate of transfecting miR-486-5p mimics was significantly higher than that of transfecting miR-486-5p mimics( P < 0. 05,P < 0. 01). Conclusion Schisandrin B can induce apoptosis of human thyroid carcinoma BCPAP cells by up-regulating miR-486-5p expression,and therefore MiR-486-5p can be used as a target of targeted therapy for human thyroid carcinoma.
