MMP-9在舌癌中的表达及其意义

【摘要】 目的研究MMP-9在舌癌(TSCC)组织中的表达及其与临床病理因素的关系。方法收集2000年1月至2010年1月广东医学院附属医院口腔科舌癌患者手术切除包埋石蜡标本60例,其中有15例取癌旁正常组织及21例有转移淋巴结,应用免疫组化PV9000法分别检测舌癌组织、癌旁组织及转移淋巴结中MMP-9的表达。结果 MMP-9在TSCC组织中的阳性表达率为78.33%(47/60),明显高于癌旁组织的20.00%(3/12),差异有统计学意义(P>0.05);在舌癌的低分化组中MMP-9阳性表达率为88.00%(22/25),明显高于高分化组的71.42%(25/35),差异有统计学意义(P>0.05);TSCC在有颈淋巴结转移组中MMP-9阳性表达率为90.48%(19/21),明显高于无颈淋巴结转移组织中的71.79%(28/39),差异有统计学意义(P>0.05);在TSCC原发灶与淋巴结转移灶中的MMP-9阳性表达率分别为78.33%(47/60)、76.19%(16/21),差异无统计学意义(P>0.05)。结论 MMP-9在TSCC组织中和有颈淋巴结转移组织中高表达,MMP-9可作为评估舌癌浸润和转移的重要指标。

【Abstract】 Objective To study the expression of matrix metalloproteinase-9(MMP-9) in tongue squamous cell carcinoma(TSCC) tissues and its relationship with clinical pathological factors. Methods A total of 60 cases of surgical resection of paraffin embedded specimens, which came from tongue cancer patients who admitted to Department of Stomatology of the Affiliated Hospital of Guangdong Medical University from January 2000 to January 2010,were collected as the research objects. At the same time, 15 cases of tumor-adjacent normal tissues and 21 cases of metastatic lymph nodes were also included in this research. The expression of MMP-9 in tongue squamous cell carcinoma, tumor-adjacent tissues and metastatic lymph node was detected by immunohistochemistry(PV9000). Results The positive expression of MMP-9 in TSCC tissues was 78.33%(47/60), which was significantly higher than 20.00%(3/12) in tumor-adjacent tissues(P<0.05). The positive expression of MMP-9 in tongue squamous cell carcinoma/low differentiation group was 88.00%(22/25), which was significantly higher 71.42%(25/35) in high differentiation TSCC group(P<0.05). The positive expression of MMP-9 in cervical lymph node metastasis was 90.48%(19/21), which was significantly higher than 71.79%(28/39) in without lymph node metastasis tissues(P<0.05). The positive expression of MMP-9 in focal TSCC in primary tumor and lymph node metastasis rate were 78.33%(47/60), 76.19%(16/21), respectively, and there was no significant difference(P>0.05). Conclusion MMP-9 has the high expression in TSCC and cervical lymph node metastasis tissues, which can serve as an important indicator to evaluate the invasion and metastasis of tongue cancer.
