香水柠檬LFB2基因的克隆及表达分析

【摘要】 无籽是香水柠檬的重要特征,自交不亲和是香水柠檬无籽的主要原因,为了深入了解香水柠檬自交不亲和的分子机理。本研究在通过De novo技术获得F-box基因片段的基础上,以香水柠檬嫩叶为材料,利用RT-PCR技术成功地克隆出长度为1 357 bp的F-box基因,命名为LFB2。生物信息学分析发现该基因编码446个氨基酸,氨基酸的C端具有两个WD结构域,属于FBXW蛋白,其蛋白分子量为111.62 kD,等电点:8.97。利用qRT-PCR分析LFB2基因在花器官不同组织的表达情况,发现该基因在雄蕊中的表达量最高,是其它花器官的十倍左右;在逆境胁迫下,该基因的表达量也有明显的变化,以上研究表明LFB2基因对花粉的发育和逆境胁迫中有一定的作用。

【Abstract】 Seedlessness is a very important characteristic of Xiangshui lemon, which is mainly caused by self-incompatibility. In order to further understand the genetic mechanism of the self-incompatibility of Xiangshui lemon, a length of 1 357 bp F-box gene was cloned by RT-PCR which was named LFB2 with lemon leaves as materials in this study on the basis of F-box gene fragment obtained by De Novo. Bioninformatics analysis showed that this gene encoded 446 amino acids, contained two WD domains at the C-terminal of the amino acid, and belonged to the FBXW protein, of which the protein molecular weight was 111.62 kD and the isoelectric point was8.97. The LFB2 expression in different flower organs was analyzed by q RT-PCR, and the results indicated that the gene was expressed highest in the stamens, which was about 10 times as much as that in other flower organs; Also,under adversity stress, the gene expression changed significantly. These results showed that LFB2 gene played a role in the growth and development of pollen and under stress environment.
