PUFA对LPS致ALI大鼠的氧化应激及TLR4、TNF-α的影响

【摘要】 目的探究并分析ω-3多不饱和脂肪酸(PUFA)对内毒素脂多糖(LPS)致急性肺损伤(ALI)大鼠的氧化应激及Toll样受体-4(TLR4)、肿瘤坏死因子α(TNF-α)的影响。方法选取健康成年Wistar大鼠120只(清洁级,体质量180~240g),按照随机数字表法将其分为对照组和观察组,每组60只,对照组大鼠又随机分为未处理组、处理后6h组(LPS注射)、处理后24h组(LPS注射),每组20只。观察组大鼠又随机分为未处理组、处理后6h组(PUFA+LPS注射)、处理后24h组(PUFA+LPS注射),每组20只。观察2组大鼠处理前、处理后6h、处理后24h超氧化物歧化酶活性(SOD)、丙二醛(MDA)、细胞间黏附分子-1(ICAM-1)、TNF-α、TLR4相对表达量。结果观察组和对照组大鼠处理前后SOD水平比较差异有统计学意义(P<0.05);观察组和对照组大鼠处理前SOD水平比较差异无统计学意义(P>0.05);随着处理时间的推移,2组大鼠SOD水平均有所降低,但观察组大鼠降低程度低于对照组,差异有统计学意义(P<0.05);2组大鼠处理前后MDA水平比较差异有统计学意义(P<0.05);随着处理时间的推移,2组大鼠MDA水平均有所升高,但观察组大鼠升高程度明显低于对照组,差异有统计学意义(P<0.05);2组大鼠处理前后ICAM-1水平比较差异有统计学意义(P<0.05);随着处理时间的推移,2组大鼠ICAM-1、TNF-α水平有所升高,但观察组大鼠升高程度明显低于对照组,差异有统计学意义(P<0.05);观察组大鼠肺组织TLR4相对表达量低于对照组,差异有统计学意义(P<0.05)。结论 PUFA能有效降低LPS致ALI大鼠的氧化应激反应及炎症水平,可能有助于ALI的治疗。

【Abstract】 Objective To explore and analyze the effects ofω-3 polyunsaturated fatty acid(PUFA)on oxidative stress and Toll like receptor-4(TLR4)and tumor necrosis factor alpha(TNF-α)in rats with acute lung injury(ALI)induced by lipopolysaccharide(LPS).Methods 120 healthy adult Wistar rats(clean grade;weight,180-240 g)were randomly divided into control group and observation group,60 rats in each group,rats in the control group were randomly divided into untreated group,6 hgroup(LPS injection),and 24 hgroup(LPS injection),20 rats in each group.The rats in the observation group were randomly divided into untreated group,6 hgroup(PUFA+LPS injection),and 24 hgroup(PUFA+LPS injection),20 rats in each group.The relative expression of Superoxide Dismutase(SOD)before treatment,6 hafter treatment and 24 hafter treatment,Malondialdehyde(MDA),intercellular cell adhesion molecule-1(ICAM-1),TNF-α,and TLR4 in two groups were observed.Results The difference of SOD levels before and after treatment in the two groups of rats was statistically significant(P<0.05),the difference of SOD levels before treatment in the two groups of rats was not statistically significant(P>0.05).As the processing time went on,the level of SOD in the two groups of rats decreased,but the decrease of the rats in the observation group was lower than that of the control group,and the difference was statistically significant(P<0.05),the difference of MDA level before and after treatment in two groups of rats was statistically significant(P<0.05).As the processing time went on,The level of MDA in the two groups of rats increased,but the increase of the rats in the observation group was significantly lower than that of the control group,and the difference was statistically significant(P<0.05).The difference of ICAM-1 level before and after treatment in the two groups of rats was statistically significant(P<0.05).As the processing time went on,the levels of ICAM-1 and TNF-alpha in the two groups increased,but the increase of the rats in the observation group was significantly lower than that in the control group,the difference was statistically significant(P<0.05).The relative expression of TLR4 in the lung tissue of the observation group was significantly lower than that of the control group(P<0.05).Conclusion PUFA could effectively reduce the oxidative stress and inflammation in ALI rats induced by LPS,which may contribute to the treatment of ALI.
