诺如病毒GⅡ.4亚型新变异株全基因组序列的研究

【摘要】 目的分析诺如病毒GⅡ.4亚型新变异株的全基因组序列,了解其变异特点。方法从264例患者腹泻物中提取RNA并进行cDNA合成,对阳性样本进行PCR鉴定,扩增片段进行测序。对测序结果显示为诺如病毒序列的进行全基因组测序并分析。结果分离到新的悉尼2012GⅡ.4变异株的类似株(荆州2013GⅡ.4株),全基因组序列测定发现新分离株出现了较大的变异,包括VP1基因高变区(P2区)的突变。结论悉尼2012GⅡ.4变异株的类似株已传播到中国,GⅡ.4变异株VP1进化较快。提醒应密切关注诺如病毒在中国的传播和进化情况,以助于疫苗的开发和治疗性单抗的制备。

【Abstract】 Objective Analysis of the complete genome sequence about the newly emerged pandemic norovirus GⅡ.4genotype,to understand its variation characteristics.Methods 264 patients were collected with diarrhea.The RNA was extracted from 264 fecal specimens and cDNA was synthetized.The positive samples were amplified by PCR,the amplified fragments were sequenced.The complete genome sequences of the norovirus was sequenced and analyzed.Results A new norovirus variant strain of Jingzhou GⅡ.4,apleiston of Sydney GⅡ.4was isolated.A large variation was found in the new variant subtype,which was mutated including in the hypervariable P2 domain of the major capsid protein VP1.Conclusion The result demonstrates the variant strain of Sydney GⅡ.4was spread to China.VP1 of norovirus GⅡ.4is evolving rapidly.The spread and evolution situation of the norovirus GⅡ.4need to be closely monitored in China for the development of effective vaccines and therapeutic monoclonal antibodies.
