氯化锂抑制铜绿假单胞菌感染引起的炎性反应调节机制研究
The modulation mechanism of LiCl inhibited the Pseudomonas aeruginosa-induced inflammation
【摘要】 目的探讨氯化锂(LiCl)在铜绿假单胞菌感染时对炎性反应的调节作用。方法 Western-blot检测LiCl应用的有效性;实时荧光定量聚合酶链式反应(qPCR)检测铜绿假单胞菌感染的巨噬细胞和中性粒细胞中炎症因子的表达;流式细胞术检测铜绿假单胞菌感染前后巨噬细胞和中性粒细胞凋亡情况。结果 Western-blot结果显示,LiCl能在巨噬细胞和中性粒细胞中促进糖原合成激酶3,9位丝氨酸的磷酸化[p-GSK-3β(Ser 9)],上调β-连环蛋白(β-catenin)的表达,证明LiCl应用的有效性;qPCR结果显示,在铜绿假单胞菌感染后,LiCl促进抑炎因子的表达,抑制促炎因子的表达;流式细胞术结果显示,在铜绿假单胞菌感染后,LiCl促进巨噬细胞和中性粒细胞的凋亡。结论在铜绿假单胞菌感染后,LiCl通过调节炎性因子的分泌和炎性细胞的凋亡发挥免疫负调节作用。
【Abstract】 Objective To explore the role of LiCl in modulating bacterial-mediated inflammation after Pseudomonas aeruginosa infection.Methods Western-blot was used to determine the efficacy of LiCl usage.The expression of inflammatory cytokines in Pseudomonas aeruginosa-infected macrophages and neutrophils was detected by qPCR.Cell apoptosis was measured by flow cytometry.Results Western-blot data showed that LiCl up-regulated the protein levels of p-GSK-3β(Ser 9)andβ-catenin in macrophages and neutrophils,indicating the efficacy of LiCl usage.qPCR data indicated that LiCl enhanced the expression of anti-inflammatory cytokines and suppressed the expression of pro-inflammatory cytokines in Pseudomonas aeruginosa-infected macrophages and neutrophils.Flow cytometry data indicated that LiCl could promoted the apoptosis of Pseudomonas aeruginosa-infected macrophages and neutrophils.Conclusion LiCl inhibited the Pseudomonas aeruginosa-induced inflammation,via regulating the inflammatory cytokine expression and the apoptosis of inflammatory cells.
