卡瓦叶子的化学成分和细胞毒活性研究
Investigation on the Chemical Constituents from the Leaves of Kava
【摘要】 选取卡瓦叶子为实验材料,通过多次硅胶柱层析、凝胶柱层析等方法对各个部位进行分离纯化,然后借助于核磁共振氢谱、碳谱、质谱等方法进行结构鉴定,最终从卡瓦叶子中分离出四个化合物,分别为:查尔酮A,查尔酮B,查尔酮C,5,7-二羟基-4’-甲氧基酮。采用MTT法,对四个化合物进行药理活性测试,结果发现,只有化合物4对细胞NCI-H460的IC50为42μg/mL,有比较弱的细胞毒性。
【Abstract】 Four compounds were obtained from the Et OAc extracted by a combination of column chromatography of silica gel, Sephadex LH-20 mamy times. Their structures were determined on the basis of 1 HNMR, 13 C NMR, 2 DNMR, UV, IR, and ESIMS spectra data and were identified as Flavokawain A(1), Flavokawain B(2), Flavokawain C(3),5,7-dihydroxy-4’-methoxyflavone(4). The pharmacological activities of four compounds were tested by MTT assay. The result showed that cytotoxicity of compound 4 were found to be weak against NCI-H460 Cells with IC50 values of 42 μgmL-1.
