鹿蹄草菌根真菌的分离与鉴定

【摘要】 为鹿蹄草耐寒性与其菌根真菌的关系研究奠定基础,对采集于黑龙江省伊春市凉水自然保护区的野生鹿蹄草根部真菌进行分离,根据形态学特征和rDNA ITS序列分析进行鉴定。结果表明:分离到的菌株分为8个形态类型且菌落颜色、质地和外形不同;菌丝特征差异较大,多具隔膜;8株菌株的生长速度不同,生长最快的菌株培养15d,菌落直径达7.8cm;最慢的培养30d仅6.5cm;rDNA ITS序列分析显示,新丛赤壳属(Neonectria)1株、木霉属(Trichoderma)1株、镰刀菌属(Fusarium)1株和Uncultured fungus 5株,其中镰刀菌属真菌在鹿蹄草根中为首次发现。鹿蹄草能与多种属真菌建立共生关系,在一定程度上体现了鹿蹄草内生真菌的生物多样性。

【Abstract】 To lay the foundation for the relationship between cold resistance and mycorrhizal fungi of P.calliantha,the fungi of wild Pyrola which collected from Liangshui Nature Reserve in Yichun City of Heilongjiang Province were isolated.Results:According to the morphological observation and rDNA ITS sequence analysis,isolation strains were separated into eight types and the color,texture and appearance of the colonies were different; The characteristics of hyphae were different,and there were many diaphragms;The growth rate of eight strains indicated the diameter of the fastest growing strain was up to7.8cm in 15 d,and the slowest culture was only 6.5cm in 30 d.In addition,rDNA ITS sequence analysis showed that they were Neonectria,Trichodermaand five Fusarium.Among themFusarium was the first time that found in the root of Pyrola.The results showed that Pyrolaand many fungal taxa to establish a symbiotic relationship,in a certain extent reflects the endophytic fungal species diversity of Pyrola.
