广谱角蛋白降解菌的筛选及其降解特性分析
A Broad-spectrum Keratin-degrading Strain:Screening and Degradation Characteristics
【摘要】 采用富集驯化的方法,筛选到一株广谱高效角蛋白降解菌株E-26。经形态观察、生理生化特性和16S rDNA基因分析,该菌株初步鉴定为蜡样芽孢杆菌。产酶特性及发酵条件优化结果表明麦芽糖和酵母粉是菌株E-26降解猪毛的最佳外加碳氮源pH 8.0、温度40℃、接种量10%、底物浓度1.0%为其降解猪毛的最适反应条件。在该优化条件下,该菌株发酵培养10 d后,最高酶活性为57.8U/mL,比初始酶活提高了37.4%,最高猪毛降解率达71.2%,比初始猪毛降解率提高了41.9%。
【Abstract】 A high-efficiency keratin-degrading strain(named E-26) was screened by enrichment cultivation and gradient acclimation.The strain was identified as Bacillus cereus on the basis of morphological examination,physiological and biochemical characterization,as well as 16 S rDNA sequence and phylogenetic analysis.The laboratory experiment results suggested,in terms of environmental factors and enzymology characteristics,that the best carbon and nitrogen source of E-26 degrading pig hairs were maltose and yeast powder;the optimum reaction condition of degrading pig hairs was:pH 8.0,temperature 40 ℃ inoculum dose 10%and substrate concentration 1%.Under the above mentioned optimized condition and fermental cultivation for 10 d,the keratinase activity of E-26 strain could reach 57.8 U/mL,37.4%higher than initial activity;and the rate of degradation of E-26 strain reached 71.2%,41.9%higher than the initial degradation rate.
