草鱼鱼鳔酶解液的抗氧化活性及理化性质研究
Research on the Antioxidant Activity and Physiochemical Properties of the Hydrolyzate of Grass Carp Bladder
【摘要】 以草鱼鱼鳔为原料,利用生物酶降解技术制备鱼鳔酶解液。以1,1-二苯基-2-苦基苯肼(DPPH)自由基清除率为试验指标,分别考察了酶种类、温度、时间、料液比、酶量等因素对酶解效果的影响。并采用高效液相色谱法(HPLC)和电感耦合等离子体质谱法(ICP-MS)分别对草鱼鱼鳔酶解液中的游离氨基酸、重金属含量进行分析。筛选出最佳酶解条件为:木瓜蛋白酶0.5%,温度55℃,酶解时间5 h,料液比50 mg/mL。在此条件下,DPPH清除率可达78.92%。结果表明,草鱼鱼鳔酶解液中呈味氨基酸甘氨酸、丙氨酸和精氨酸含量较高,重金属含量远低于现行《国家标准保健食品》的重金属检出限。
【Abstract】 Thehydrolyzateof grass carp bladder was prepared by using the technology of biological enzyme degradation.The effects of enzyme type, temperature, time, liquid ratio and enzyme on the enzymolysis were investigated by using the free radical scavenging rate of 1,1-diphenyl-2-bitteryl phenylhydrazine(DPPH) as the experimental index. The contents of free amino acids and heavy metals in the enzymolysic solution of grass carp were studied by high performance liquid chromatography(HPLC) and inductively coupled plasma mass spectrometry(ICP-MS). The best enzymolysic conditions were screened out as follows: 0.5% of papain, temperature of 55℃, 5 hours of enzymolysis time, liquid ratio of 50 mg/ml. Under these conditions, DPPH clearance rate reached as high as 78.92%, which showed that the contents of amino acid glycine, alanine and arginine were comparatively higher in the enzymolysis solution of grass carp, and the content of heavy metal was much lower than that of "national standard health food".
