车前草中苯乙醇苷分离纯化研究
Study on separation and purification of phenylethanoid glycosides from Plantago Herba by macroporous resin
【摘要】 目的:探讨大孔树脂分离纯化车前草中苯乙醇苷类成分的工艺。方法:以苯乙醇苷量为指标,考察HPD100、HPD400、XDA-1、D101、LX-T28和AB-8对苯乙醇苷的吸附性能,并系统考察分离纯化苯乙醇苷类成分的最佳工艺条件。结果:以D101树脂为吸附剂,上样液中苯乙醇苷类含量1.3-1.5mg/m L,体积流量为2BV/h,p H调节至5左右,氯化钠浓度约为1.5mol/L,充分吸附后,先用10BV水洗至流出液无色,随后用8BV 10%乙醇除杂,最后用5BV 50%乙醇洗脱,洗脱液减压浓缩干燥,所得提取物中苯乙醇苷含量为65.5%。结论:D101大孔树脂可有效分离纯化车前草提取物中的苯乙醇苷类成分,且工艺稳定、重现性好。
【Abstract】 Objective: To study the separation and purification of phenylethanoid glycosides from Plantago Herba by macroporous resin.Methods: The adsorption properties of HPD100,HPD400,XDA-1,D101,LX-T28 and AB-8 were investigated by using the amount of benzene as an index,and the optimum conditions for the separation and purification of the glycosides were investigated.Results: The use of D101 resin as adsorbent,the sample content of phenylethanoid glycosides was 1.3-1.5 mg/m L,volume 2 BV/h,p H adjusted to 5,the concentration of sodium chloride is about 1.5 mol/L,fully after adsorption,first with 10 BV water outflow to colorless,followed by 8 BV 10% ethanol impurity,finally using 5 BV 50% ethanol,eluent concentrate drying,phenylethanoid glycosides content in the extract was 65.5%.Conclusion: D101 macroporous resin can be effectively isolated and purified from the extract of Plantago Herba,and the process is stable and the reproducibility is good.
