基于灰色关联分析的黄连谱效关系研究

【摘要】 目的:明确黄连超高效液相色谱-质谱联用技术(UPLC-MS/MS)指纹图谱与其抗炎药效的相关性,为黄连质量控制和药效物质基础研究提供科学依据。方法:采用灰色关联度分析法研究黄连UPLC-MS/MS指纹图谱和黄连对湿热型溃疡性结肠炎大鼠模型血清白细胞介素4(IL-4)影响的谱效相关性。结果:建立了黄连UPLCMS/MS指纹图谱的共有模式,确定了能够表征黄连特征的13个共有峰。与模型组比较,不同批次的黄连给药组均能不同程度的升高湿热型溃疡性结肠炎大鼠模型血清IL-4的含量(P<0.05,P<0.01)。黄连UPLC-MS/MS指纹图谱的各共有峰与其抗炎药效作用的关联度均>0.6。结论:通过谱效关系研究明确了黄连发挥抗炎药效作用是盐酸小檗碱、盐酸药根碱、黄连碱等多成分共同作用的结果。

【Abstract】 Objective: To determine the relation between UPLC-MS/MS fingerprint andits anti-inflammatory effect to provide scientific evidence for the study of the quality control and pharmacodynamic material basis of Coptidis Rhizoma.Methods: The spectrum-effect relationship study was performed by using the method of grey correlation degree to acquire the correlation between UPLC-MS/MS fingerprint of Coptidis Rhizoma and the information of its influence on the serum IL-4 content of rat model of ulcerative colitis with damp-heat syndrome.Results: The common pattern of UPLC-MS/MS fingerprint of Coptidis Rhizoma was established with confirming 13 common peaks.Compared the model group,the drug groups of different batches Coptidis Rhizoma increased the serum IL-4 content of rat model of ulcerative colitis with damp-heat syndrome(P<0.05,P<0.01).The correlation degree between the common pattern of UPLC-MS/MS fingerprint of Coptidis Rhizoma and its anti-inflammatory effect was over 0.6.Conclusion: The spectrum-effect relationship indicated that the anti-inflammatory effect of Coptidis Rhizoma is originated from the combination action of multicomponent including berberine hydrochloride,jatrorrhizine hydrochloride,and coptisine.
