蚂蟥酶解物不同部位的抗凝活性研究

【摘要】 目的:研究蚂蟥酶解物不同部位的抗凝活性。方法:分别采用有机溶剂沉淀法、超滤法以及C18反相色谱法将蚂蟥酶解物分为不同有机溶剂沉淀部位、大分子和小分子部位、强极性和弱极性部位,以凝血酶时间为抗凝活性指标测定各部位的抗凝活性;同时采用外翻肠囊模型,研究不同部位经大鼠肠吸收后的抗凝活性差异。结果:利用有机溶剂沉淀法所得各部位的中、高剂量组均有抗凝活性(P<0.05);分级超滤所得分子量>10KD的部位具有显著抗凝活性(P<0.01),却不易被肠吸收,分子量<10KD的部位虽然抗凝活性较低(P<0.05),但较易通过肠黏膜;强极性部位和弱极性部位均有抗凝活性(P<0.05,P<0.01),但强极性部位活性更显著,更易透过肠黏膜。结论:蚂蟥酶解物活性分散,成分复杂,其发挥抗凝作用的成分可能为一些小分子大极性多肽类成分。

【Abstract】 Objective: To study the anticoagulant activity of different fractions from enzymatic hydrolysis of Whitmaniapigra Whitman. Methods:The organic solvent precipitation and ultrafiltration method, reverse phase chromatography were used to separate the enzymolysis product of Whitmania pigra Whitman into different parts, including fraction of organic solvent precipitation, fraction of different molecules, and fraction of different polarity. Thrombin time(TT) was chosen as an activity index to measure the anticoagulant activity of different fractions. In the meantime, the methodology of everted sacs was used to investigate the absorption characteristics of different fractions in rat intestine. Results: Each parts obtained from organic solvent precipitation had some anticoagulant activity(P<0.05). Each fractions with different molecular weight isolated by ultrafiltration method had obvious anticoagulant activity, especially for the fraction of MW>10KD(P<0.01). However, compared with the components of MW<10KD, the former showed poor anticoagulant activity after intestinal absorption(P<0.05). Each fraction of different polarity had some anticoagulant activity(P<0.05, P<0.01). The fraction with strong polarity showed better anticoagulant activity before and after intestinal absorption than the weak polarity fraction. Conclusion: The anticoagulant active peptides in enzymatic hydrolysis of Whitmania pigra Whitman were decentralized and sophisticated, and its material base of anticoagulation maybe some peptides with low molecular and strong polar.
