基于数据挖掘和网络药理学的儿童食积胃肠积热证组方用药规律及作用机制分析

【摘要】 目的:挖掘儿童食积胃肠积热证的组方用药规律,分析处方可能存在的药物靶标及相互作用,探索核心药物的作用机制。方法:收集CNKI中关于儿童食积胃肠积热证的病例报道文献,采用TCMISS软件分析药物间的关联规则,挖掘核心药物及新处方;运用SPSS软件对药物的性味归经进行系统聚类分析;运用TCMSP、String及KEGG数据库分析新处方各药物靶标间相互作用关系及相关通路。结果:在筛选出的150首处方中,挖掘出6个核心药物组合和3首新处方,总结出性味归经相近的9个高频药群,得到新处方中相关药物靶标330个,这些靶标构成的网络中共存在相互作用2 961项、相关通路137条(FDR<0.01),其中与消化系统关系最密切的7条。结论:治疗儿童食积胃肠积热证以消积导滞、理气健脾、清热除湿为主,其机制可能通过调控消化液的分泌,调控肠道消化及免疫、机体对微生物的免疫功能等多靶标复杂通路的共同干预实现。

【Abstract】 Objective: To explore the medication rules of herbal prescriptions for children indigestion with syndrome of retention of heat in stomach, and analyze the possible drug targets and interactions, in order to explore the mechanism of the core drugs. Methods: The prescriptions for treating children indigestion with syndrome of retention of heat in stomach were collected from CNKI, the association rules of herbs were analyzed by using TCMISS software, the core components of herbs and new formulas were mined, and the properties, flavors and meridian tropism of herbs were analyzed by using systematic cluster analysis method with SPSS software. The interactions between targets and the related signaling pathways of new prescriptions were analyzed by Traditional Chinese Medicine Systems Pharmacology Database(TCMSP), Search Tool for the Retrieval of Interacting Genes/Proteins(STRING) and Kyoto Encyclopedia of Genes and Genomes(KEGG). Results: Among the 150 screening prescriptions, 6 core drug combinations and 3 new prescriptions were obtained, 9 high-frequency herbs groups of similar properties, flavor and meridian tropism and 330 related targets. In the network comprised with those action targets, there were 2 961 actions and 137 signaling pathways(FDR<0.01). Among those signaling pathways, there were 7 pathways had the most closely relationship with the digestive system. Conclusion: For children with indigestion due to accumulated heat in stomach, the commonly used therapy of TCM were ridding of retained food, regulating qi and tonifying the spleen and clearing heat and dampness. And its action was achieved by the regulation of digestive juice secretion, regulation of intestinal digestion and immunity, the body’s immune function of microorganisms and other multi-target complex pathway.
