ALA-PDT对食管癌Eca-109凋亡相关基因表达的影响

【摘要】 目的研究凋亡相关基因Bak-1、FADD在食管癌Eca109细胞ALA-PDT中的作用及初步探讨其在食管癌ALA-PDT凋亡通路中的作用。方法实验分为对照组和治疗组,治疗组细胞给予ALA孵育6 h后在630 nm Diomed光动力治疗系统下分别光照0.5、3、24 h后收集细胞,采用real-time PCR法检测Bak-1、FADD基因的表达。结果 ALA-PDT作用3、24 h后,FADD的相对表达量均较对照组降低(P﹤0.05);ALA-PDT作用0.5、3 h后,Bak-1的相对表达量低于对照组,差异有统计学意义(P﹤0.05);但FADD的相对表达量在作用0.5 h后,Bak-1的相对表达量在作用24 h后与对照组相比,差异均无统计学意义(P﹥0.05)(P﹥0.05)。结论 Fas通路可能为ALAPDT诱导Eca-109细胞凋亡的通路之一,FADD可能为该凋亡通路的靶基因之一。
【Abstract】 Objective To investigate the role of apoptosis related gene Bak-1 and FADD in ALA mediated photodynamic therapy(ALA-PDT) in esophageal cancer cell line Eca-109 and explore its role in apoptosis pathway in PDT.Method The esophageal cancer cell line was divided as control group or study group,and the cells in study group were incubated with ALA for 6 h,and then were irradiated using the 630 nm Diomed laser system for 0.5 h,3 h and 24 h before harvest,and then quantitative real-time polymerase chain reaction(q RT-PCR) assay was used to detect the gene expression level of Bak-1 and FADD.Result The relative expression of FADD in Eca-109 cells were significantly down-regulated at 3 h and 24 h after the ALA-PDT treatment(P<0.05),and the relative expression of Bak-1 were down-regulated at 0.5 h and 3 h after treatment compared with that of control group,but the differences were not significant after treatment for 0.5 h in FADD and 24 h for Bak-1 gene(P>0.05).Conclusion Fas pathway may be involved in the apoptosis pathway induced by ALA-PDT in Eca-109 cell,and FADD may be the target gene of the pathway.
