消癥汤对膀胱肿瘤T24细胞凋亡及相关蛋白Slit2、Bim、Bad的影响

【摘要】 目的探讨消癥汤抑制膀胱肿瘤复发的可能作用机制。方法取对数生长期膀胱肿瘤T24细胞,调整浓度为1×104个/ml,每孔200μl接种于96孔板中,常规培养24 h后每孔加入200μl含有不同浓度消癥汤(药物终浓度分别为5、10、20、40、80、160、320、640、1280μg/ml)的完全培养基,每种浓度设5个平行孔,干预24 h后测定消癥汤对T24细胞增殖抑制作用,并计算消癥汤半数抑制浓度(IC50)。根据结果取低、中、高剂量的消癥汤培养T24细胞24 h,并设0μg/ml消癥汤做对照,检测消癥汤对T24细胞凋亡的影响,并测定低、中、高剂量组Slit2、Bim、Bad蛋白及基因表达。结果消癥汤对T24细胞的IC50为127.14μg/ml,中剂量取120μg/ml,低剂量取60μg/ml,高剂量取240μg/ml。对照组T24细胞凋亡细胞百分比为(4.41±0.50)%,低剂量组为(4.47±0.13)%,中剂量组为(11.04±1.32)%,高剂量组为(12.08±0.55)%。与对照组比较,中剂量组和高剂量组凋亡细胞百分比明显升高(P<0.05或P<0.01)。与低剂量组比较,中、高剂量组能够增加与凋亡相关的Slit2、Bim、Bad蛋白及基因表达(P<0.05)。结论消癥汤能抑制膀胱肿瘤T24细胞的增殖,可能与增加凋亡相关蛋白Slit2、Bim、Bad的表达有关。

【Abstract】 Objective To explore the possible mechanism of Xiaozheng Decoction( 消癥汤) inhibiting recurrence of bladder cancer. Methods Bladder cancer T24 cells of logarithmic phase were extracted. The concentration after regulating was 1 × 104 cells / ml. The cells were inoculated in 96-well plates,200μl in each pore. After 24 hours of routine culture,complete medium 200 μl containing Xiaozheng Decoction in different concentration( with final concentration of drug 5,10,20,40,80,160,320,640,or 1280μg / ml) was added to each pore. Five parallel pores of each concentration were designed. After intervention for 24 hours,the inhibition effect of Xiaozheng Decoction on T24 cell multiplication was examined with methyl thiazolyl tetrazolium( MTT) method. The Xiaozheng Decoction concentration of median lethal dose( IC50) was calculated. According to the results,T24 cells were cultured with low,medium and high dose of Xiaozheng Decoction for 24 hours. 0 μg / ml of Xiaozheng Decoction was designed as control. The impact of Xiaozheng Decoction on T24 apoptosis was examined with Annexin V / propidium iodide( PI) double staining method. The expressions of Slit2,Bim,and Bad proteins and genes in the low,medium and high dose groups were detected with Western blot and reverse transcriptase-polymerase chain reaction( RT-PCR) methods. Results IC50 of Xiaozheng Decoction to T24 cells was 127. 14μg / ml,120μg / ml of medium dose,60μg / ml of low dose,and240μg / ml of high dose. The percentage of apoptotic cells of T24 cells in the control group was( 4. 41 ± 0. 50) %,( 4. 47 ± 0. 13) % in the low dose group,( 11. 04 ± 1. 32) % in the medium group,and( 12. 08 ± 0. 55) % in the high dose group. Compared with the control group,the percentage of apoptotic cells in the medium and high dose groups increased( P < 0. 05 or P < 0. 01). Compared with the low dose group,the expressions of Slit2,Bim,and Bad proteins and genes which were related to apoptosis in the medium and high dose groups increased( P < 0. 05).Conclusion Xiaozheng Decoction might inhibit the proliferation of bladder cancer T24 cells. It might be related to increasing the expressions of apoptosis related proteins,such as Slit2,Bim and Bad.
