灯盏花素对糖尿病大鼠肾脏VEGF表达的影响

【摘要】 目的:观察灯盏花素(breviscapine)对链脲佐菌素(STZ)诱导的糖尿病大鼠肾脏VEGF表达的影响。方法:大鼠尾静脉注射STZ 45mg/kg建立糖尿病大鼠模型,将成模糖尿病大鼠随机分为糖尿病模型组、灯盏花素治疗组(10mg/kg剂量组;15mg/kg剂量组;20mg/kg剂量组),同时设立正常对照组。治疗8w后检测各组大鼠血糖(FBG)、肾指数(肾重/体重)、肌酐(SCr)、尿素氮(BUN)、尿白蛋白排泄率(UAER)、光镜和电镜观察肾皮质形态学改变,免疫组化法和Western blot法检测VEGF定位和半定量表达。结果:与糖尿病模型组比较,3种不同剂量灯盏花素治疗组肾指数、UAER、s Cr、BUN明显降低,光镜及电镜下肾脏病理改变有明显改善,肾组织VEGF表达减少。结论:灯盏花素对糖尿病大鼠肾脏具有一定的保护作用,可能与其抑制糖尿病大鼠肾脏VEGF表达有关。

【Abstract】 Objective: To observe effects of breviscapine Injection on renal function and expressions of VEGF in kindey of diabetic rats induced by STZ. Methods: SD rats were created the experimental model of diabetes by injecting STZ through vena caudalis. They were randomly divided into DM group and breviscapine groups( 10 mg / kg,15 mg / kg,20 mg / kg). Other rats were set as normal control group. Eight weeks later,blood glucose,renal index,SCr,BUN,UAER were assayed. The morphologicai change of renal cortex was observed by light microscope and electron microscope. Immunohistochemistry method and western blot were used to determine the expressions of VEGF in renal tissue. Results: Compared with DM group,in Bre groups,the blood glucose level was unchanged,whereas renal index and UAER were decreased,and SCr,BUN were improved obviously( P < 0. 01 or P < 0. 05). Kindey structure was improved obviously too. Moreover,expressions of VEGF in renal tissue of diabetic rats in Bre groups were much less than those in DM group( P < 0. 01). Conclusion: Breviscapine has protective effects on kindey in diabetic rats,which may be associated with lessening expressions of VEGF in renal tissue.
