陈皮姜中橙皮苷、川陈皮素和6-姜酚的药物动力学分析
Pharmacokinetic analysis of hesperidin,nobiletin and 6-gingerol of Chenpi-Jiang
【摘要】 目的:建立一个简单、灵敏、快速的高效液相色谱法测定陈皮姜中橙皮苷、川陈皮素和6-姜酚在大鼠体内的血药浓度,并研究其药物动力学。方法:HPLC定量检测血浆中待测药物浓度,使用C18色谱柱,以甲醇-1%冰醋酸水溶液为流动相,梯度洗脱;检测波长:283 nm,328 nm;流速为1.0 ml/min;柱温30℃。结果:内源性杂质在出峰时间对样品和内标测定无干扰。橙皮苷在0.2~3.2μg/ml,川陈皮素在0.2~3.2μg/ml,6-姜酚在0.1~1.6μg/ml线性关系良好。日内、日间精密度(RSD)均小于15%,样品的24 h稳定性、长期稳定性和反复冻融稳定性良好。方法学考察均符合生物样品分析的要求。橙皮苷、川陈皮素和6-姜酚的t1/2(β)值分别是11.55±2.11,15.07±2.94,15.40±2.14;Cmax值是:1.88±0.37,2.55±0.42,0.98±0.26;AUC0→∞值是:16.59±2.20,16.90±3.91,3.56±0.96。结论:所建立的方法简便、灵敏、重复性好、分析速度快,可用于血浆样品中橙皮苷、川陈皮素和6-姜酚的血药浓度测定和大鼠体内药物动力学研究。
【Abstract】 Objective: To develop a simple sensitive and rapid method for determining the concentrations of hesperidin,nobiletin and 6-gingerol in rat plasma by using liquid chromatography and then to investigate the pharmacokinetic profile of them in rats with the method. Methods: The Plasma concentrations of components under tested were quantified by HPLC. After a pretreatment for plasma sample,chromatographic separation was achieved on a C18 reversed phase column with mobile phase of methanol and 1% glacial acetic acid water solution by gradient elution. The flow rate was 1. 0 ml / min. The detection wavelength were set at 283 nm and 328 nm with the column temperature was set at 30 ℃.Results: There were no interference peaks co-eluting with the compounds during the retention time for hesperidin,nobiletin,6-gingerol and IS. A good linearity was obtained for hesperidin in the range of 0. 2 ~ 3. 2,nobiletin in the range of 0. 2 ~ 3. 2 and 6-gingerol in the range of0. 1 ~ 1. 6 μg / ml. The intra-day and inter-day precision was less than 15%. The Short-term stability for 24 h,Long-term stability for 30 days at-80 ℃ and freezing-thaw stability of the samples were satisfactory. The method validation was subjected to the requirements of biological sample analysis. The t1 /2( β),Cmax and AUC0→∞of hesperidin,nobiletin,6-gingerol are 11. 55 ± 2. 11,1. 88 ± 0. 37,16. 59 ± 2. 20 and15. 07 ± 2. 94,2. 55 ± 0. 42,16. 90 ± 3. 91 and 15. 40 ± 2. 14,0. 98 ± 0. 56,3. 56 ± 0. 96. Conclusion: The established method is an sensitive,reliable and specific method for the pharmacokinetic study of hesperidin,nobiletin and 6-gingerol of Chenpi-Jiang( 陈 皮姜) in rat plasma.
