MTHFR基因G1793A多态性与子痫前期发生的相关研究

【摘要】 目的研究亚甲基四氢叶酸还原酶(MTHFR)基因G1793A多态性与子痫前期发生的关系。方法采用聚合酶链反应—限制性片段长度多态性(PCR-RFLP)法对108例妊娠高血压疾病子痫前期患者,120例正常妊娠女性MTHFR的G1793A进行基因分析,比较上述各组基因型和等位基因频率分布有无差异。结果 MTHFR基因G1793A突变型等位基因(A)频率在实验组和对照组中有显著性差异,GG、AA基因型频率分布差异有显著性(P<0.05)。AA基因型比GG基因型发生妊娠高血压疾病风险高3.63倍。结论 MTHFR G1793A基因多态性与妊娠高血压疾病子痫前期发生相关,AA基因型增加了妊娠高血压疾病子痫前期的发生风险,GG基因型是降低妊娠高血压疾病子痫前期发生的保护性因素。

【Abstract】 Objective:To explore the relationship between genetic polymorphisms in methylenetetrahydrofolate reductase(MTHFR),a central enzyme in folate metabolism that affects DNA methylation and synthesis, and the risk of Preeclampsia. Methods:Genomic DNA was isolated from the peripheral lymphocytes of 108 patients with Preeclampsia and 120 age matched control subjects. Polymerase chain reaction and restriction fragment length polymorphism were used to examine the polymorphisms of MTHFR genes G1793 A and the relation between these genotypes and risk of Preeclampsia was analyzed. Results:There was obvious difference between experiment and control group in MTHFR gene G1793 A mutation allele(A)frequency(P<0.05),AA and GG had obvious difference(P<0.05). The individuals with 1793 AA genotype had a 4.33 fold increased risk of developing preeclampsia compared with those who had 1793 GG genotype. Conclusions:MTHFR gene mutation allele is related to preeclampsia,AA gene mutation type increases the risk of Gestational hypertension and GG type perhaps is a protective factor in decreasing the risk of preeclampsia.
