电针对吗啡戒断大鼠VTA和NAc脑区CaMKⅡα亚基mRNA表达的影响
Effect of Electroacupuncture on the Expression of CaMKⅡα Subunit mRNA in VTA and NAc Brain Regions of Morphine Withdrawal Rats
【摘要】 目的:通过观察吗啡戒断大鼠VTA和NAc脑区CaMKⅡα亚基mRNA表达的变化,探讨电针对吗啡戒断反应的影响。方法:清洁级雄性SD大鼠56只,采用随机数字表法分对照组、模型组、针刺组和电针组,每组14只,采用剂量递增法制备吗啡依赖动物模型,给予颈背部皮下注射吗啡20 mg/kg、30mg/kg、40 mg/kg、50 mg/kg、50 mg/kg连续5日给药,第6日给予纳洛酮催促戒断,第7日开始针刺干预治疗。针刺组选取百会穴、神庭穴、肾俞穴单纯针刺,留针15 min,1次/d,连续6 d。电针组选穴同针刺组,给予电针疏密波(频率为2/100 Hz),留针15 min,1次/d,连续6 d。对照组和模型组不予任何治疗,只予捆绑固定。针刺疗程结束后新鲜冰冻取脑组织VTA和NAc脑区,采用Real Time PCR法测定其CaMKⅡα亚基mRNA的表达。结果:与对照组比较,模型组大鼠VTA和NAc脑区神经元CaMKⅡαmRNA表达显著升高,差异具有统计学意义(P<0.01)。与模型组比较,针刺和电针干预后吗啡戒断大鼠NAc神经元CaMKⅡαmRNA表达显著降低,差异具有统计学意义(P<0.05)。与针刺组比较,电针组大鼠VTA和NAc神经元CaMKⅡαmRNA表达降低,差异具有统计学意义(P<0.05)。结论:电针能够抑制吗啡戒断模型大鼠VTA和NAc脑区CaMKⅡα亚基mRNA表达,这可能是电针改善吗啡戒断反应的重要分子机制。
【Abstract】 Objective: To investigate the effect of electroacupuncture on the expression of CaMKIIα subunit mRNA in VTA and NAc brain regions of morphine withdrawal rats. Methods: 56 male SD rats were randomly divided into blank group,model group,acupuncture group and electroacupuncture group,with 14 rats in each group. Morphine dependent model was induced by subcutaneous injection of morphine in the back of the neck with dose-increasing method,and naloxone precipitated withdrawal was administered immediately after 5 days. In the acupuncture group and the electroacupuncture group Baihui( GV20),Shenting( GV24) and Shenshu( GV4) points were selected for acupuncture and the acupuncture was based on the connectionⅡ KWD808 pulse therapeutic apparatus,with 15 minutes a day,1 time a day,continuous intervention for 6 days. The expression of CaMKⅡα subunit mRNA in brain tissues of VTA and NAc was determined by Time PCR Real method. Results: Compared with the control group,the expression of CaMKⅡα mRNA in the VTA and NAc neurons in model group was significantly increased,and the difference was statistically significant( P < 0. 01).Compared with model group,the expression of CaMKⅡα mRNA in NAc neurons of morphine withdrawal rats was significantly decreased after acupuncture and electroacupuncture intervention,and the difference was statistically significant( P < 0. 05,P < 0. 01). Compared with the acupuncture group,the expression of CaMKⅡα mRNA in VTA and NAc neurons in the electroacupuncture group was decreased,and the difference was statistically significant( P < 0. 05). Conclusion: Electroacupuncture can inhibit the expression of CaMKⅡα mRNA in VTA and NAc brain regions of morphine withdrawal rats,which may be an important molecular mechanism of electroacupuncture for alleviating morphine withdrawal response.
