益心血脂康胶囊对动脉粥样硬化模型大鼠的作用
Effect of Yixin Xuezhikang Capsules on Experimental Atherosclerosis in Rats
【摘要】 目的:观察益心血脂康(YXK)对AS模型大鼠的影响。方法:SD大鼠随机分为空白对照组、模型对照组、YXK低、高剂量组和阿乐对照组。空白对照组动物摄食正常颗粒饲料,模型对照组及YXK各组在摄食高脂饲料的基础上采用ip VD3加激发免疫反应的方法复制AS模型。各组动物连续灌胃90 d,检测血清TC、TG、LDL-C、HDL-C、Ca、SOD、MDA、NO、NOS、ET、IL-6、IL-18、CPR、TNF-α、s CD40L及APN并计算动脉硬化指数(AI)和脏器指数;取胸主动脉HE染色,进行病理组织学观察。结果:与空白对照组相比,模型对照组大鼠肝脏指数、AI、血清TC、LDL-c、Ca、MDA、ET、IL-18、CPR、TNF-α和s CD40L显著升高(P<0.05),而SOD、NO、NOS和APN均有不同程度降低。与模型对照组比较,YXK组可降低AS大鼠肝脏指数、AI、血清TC、LDL-c、MDA、TNF-α、IL-6、IL-18和s CD40L水平(P<0.05),并显著提高血清NO、NOS和APN水平(P<0.05)。HE染色显示模型对照组主动脉可见内皮损伤,有粥样斑块形成,给药各组均可减少其主动脉损伤。结论:YXK通过改善AS模型大鼠脂质代谢、减少氧自由基损伤以及调节相关细胞因子的分泌或释放等功效,干预或减缓大鼠动脉粥样硬化的发生发展。
【Abstract】 Objective : To investigate the antiatherogenic effect and possible mechanisms of Yinxinkang( YXK) capsules.Methods : SD rats were randomly divided into 5 groups: normal control group,model control group,YXK capsule high and low dose groups and positive control group. Normal control group was given saline and basic food. In the rest of four groups rats were injected with VD3 and ovalbumin,while fed with high fat diet. Each group was given the corresponding drugs for 90 days. Blood serums were analyzed for TC,TG,LDL- c,HDL- c,Ca,SOD,MDA,NO,NOS,ET,IL- 6,IL- 18,CPR,TNF- α and s CD40 L. Visceral index and arteriosclerosis index( AI) were counted and the pathological observation of aorta was carried out. Results: The liver index,AI,TC,LDL- c,Ca,MDA,ET,IL- 18,CPR,TNF- αand s CD40 L were significantly higher while the SOD,NO,NOS and APN were less in model control group than those in normal control group( P < 0. 05). Compared with those in model group,the liver index,AI,TC,LDL- c,MDA,TNF- α,IL- 6,IL- 18 and s CD40 L levels were reduced,while the serum NO,NOS and APN levels were raised in the YXK capsule group. HE stain showed aortic endothelial injury and atherosclerotic plaque formation in model group and the different dose groups could restrain different degrees of aortic lesion. Conclusion: The YXK capsules can improve metabolism of lipid in model rats. It can reduce oxygen free radical damage and regulate vascular active substances. It has an interventive effect on atherosclerotic plaque formation of model animals at multiple targets.
