复方夏枯草提取物对自发性高血压大鼠的降压作用

【摘要】 目的:探讨复方夏枯草提取物对自发性高血压(SH)大鼠的降压作用。方法:将40只SH大鼠随机分为模型组、复方夏枯草提取物高、中、低剂量组、复方罗布麻片Ⅰ组,每组8只,采用无创尾动脉测压法观察复方夏枯草提取物对SHR大鼠收缩压(SBP)、舒张压(DBP)的影响,给药8周后测定血清中一氧化氮(NO)、血管紧张素Ⅱ(AngⅡ)、内皮素-1(ET-1)和心钠素(ANP)的含量,观察SHR大鼠肾脏HE染色病理变化。结果:与模型组比较,复方夏枯草高、中剂量组和复方罗布麻片Ⅰ组同期血压明显降低(P<0.05)。与模型组比较,复方夏枯草高、中剂量组和复方罗布麻片Ⅰ组血清AngⅡ、ET-1水平明显降低(P<0.01),血清NO、ANP含量明显升高(P<0.01)。病理检查提示,模型组比各给药组病变明显加重,出现肾小球肾小管萎缩、肾小球血管管壁增厚及肾小管上皮细胞变性坏死等病理改变。结论:复方夏枯草提取物具有一定的降血压作用,其机制可能与降低AngⅡ和ET-1含量、升高NO和ANP含量有关。

【Abstract】 Objective:To study the antihypertensive effect of the extract from compound Prunella vulgaris L.in spontaneous hypertension(SH) rats.Methods:Forty SH rats were randomly divided into the model group,high,middle and low dose groups of extract from vompound Prunella vulgaris L.,the compound kendir leaves tablets Ⅰ group with 8 ones in each.Non-invasive blood pressure measurement was used to detect the SBP and DBP of the SHR rats.Then the serum NO,AngⅡ,ET-1 and ANP content were measured after the eight-week treatment.The pathological changes were observed after kidney HE staining in the SH rats.Results:Compared with that in the model group,the blood pressure in high-dose treatment group,middle-dose treatment group and the positive model group was significantly decreased(P < 0.05).The AngⅡ and ET-1 levels in high-dose treatment group,middle-dose treatment group and the positive model group were decreased(P < 0.01),and NO and ANP contents in serum were significantly increased when compared with those in the model group(P < 0.01).The pathological examination showed that the pathological changes in the model group were faster than those in all the drug-treatment groups,the pathological changes included glomerular and renal tubular atrophy,glomerular vascular wall thickening and renal tubular epithelial cell degeneration or necrosis.Conclusion:The extract from compound Prunella vulgaris L.can reduce blood pressure of SH rats.The mechanism may be associated with the level reduction of Ang II and ET-1 and content elevation of NO and ANP.
