益肾化湿颗粒对糖尿病肾病小鼠足细胞保护机制研究
Protective mechanism of Yishenhuashi particle on podocytes in diabetic nephropathy mice
【摘要】 目的观察益肾化湿颗粒对糖尿病肾病(DN)小鼠肾脏足细胞裂孔隔膜蛋白(Nephrin、Podocin)、血小板衍生生长因子受体β(PDGFRβ)表达的影响,探讨其防治糖尿病肾病的机制。方法通过高脂膳食建立DN小鼠模型,随机分为正常组、模型组、益肾化湿治疗组、缬沙坦组,并每周称重。干预4周后处死小鼠,分别检测各组24 h尿蛋白量、血糖、肌酐,光镜观察肾脏病理变化,免疫荧光染色检测肾脏PDGFRβ的表达,Western Blot检测Nephrin、Podocin、PDGFRβ的表达。结果与模型组相比,益肾化湿治疗组小鼠血糖降低[模型组(37.04±2.72)mg/d L;益肾化湿组(32.51±4.65)mg/d L],尿微量白蛋白肌酐比降低[模型组(4.95±1.36)μg/mg;益肾化湿组(1.41±0.17)μg/mg],肾脏病理变化有明显改善,Nephrin、Podocin的表达显著增高(P<0.01),PDGFRβ的表达显著降低(P<0.01)。结论益肾化湿颗粒通过上调足细胞Nephrin、Podocin及降低肾脏PDGFRβ的表达,减轻DN小鼠尿蛋白排泄,具有肾功能保护作用。
【Abstract】 Objective To observe the effect of Yishenhuashi particle on the expression of Nephrin,Podocin and PDGFRβ in diabetic nephropathy( DN) mice and explore its mechanism of prophylaxis and treatment of DN. Methods DN model was established by high fat diet( HFD),randomly divided into 4 groups: normal control group,HFD group,Yishenhuashi group,valsartan group,and weighed every week. All mice were sacrificed at week 8 after the treatment. The24 h urine protein,blood sugar,creatinine were detected. The pathological changes of the glomerulus were observed with the optical microscope. The expression of PDGFRβwas detected by immunoflurescence technique and Western blot analysis. The expression of Podocin and PDGFRβwere detected by Western blot analysis. Results Compared with the HFD group,the blood sugar [HFD group( 37. 04 ± 2. 72) mg / d L & yishenhuashi group( 32. 51 ± 4. 65) mg / d L]and the microalbuminuria of the yishenhuashi group[HFD group( 4. 95 ± 1. 36) μg / mg & yishenhuashi group( 1. 41 ± 0. 17) μg /mg]were decreased,the pathological changes of the glomerulus were ameliorated,the expression of Nephrin and Podocin in podocyte were increased( P < 0. 01) and the expression of PDGFRβwere decreased( P < 0. 01). Conclusion Yishenhuashi can alleviate urine protein excretion of DN mice by upregulating the expression of Nephrin and Podocin in podocytes and inhibiting the expression of PDGFRβin kidney,having protective effect of renal function.
