一种病证结合痰浊阻窍证老年期痴呆大鼠模型的建立

【摘要】 目的:建立一种病证结合的痰浊阻窍证老年期痴呆大鼠模型。方法:61只大鼠随机分为A空白对照组、B老年性痴呆组、C血管性痴呆组、D痰浊阻窍证组、E病证结合(AD+VD+痰浊阻窍证)组,从Morris水迷宫、血脂、海马神经元超微结构等不同方面,对痰浊阻窍证老年期痴呆大鼠模型的建立进行初步探索。结果:造模成功后,Morris水迷宫实验显示B、C、E组与A组间逃避潜伏期及目标象限停留时间、原平台区穿越次数数据,差异有统计学意义。血脂化验显示,D组及E组的TC、LDL-C升高为其余各组的2倍以上。透射电镜下观察大鼠海马CA1区超微结构,可见E组的神经元细胞超微结构受损最为严重,B组次之,再次为C组与D组。结论:该模型可作为痰浊阻窍证老年期痴呆模型使用,用于老年期痴呆的实验研究。

【Abstract】 The syndrome of phlegm obstructing orifices of senile dementia rat model was established by a combination of disease and syndrome. Methods: 61 rats were randomly divided into A blank control group,B AD group,C VD group,D phlegm blocking orifices group,E combination of disease and syndrome( AD + VD + syndrome of phlegm obstructing orifices) group,from the behavior of rats,biochemical,morphological aspects preliminary exploration of establishing model,the syndrome of phlegm obstructing orifices of senile dementia rats. Results: After the success of modeling,Morris water maze test showed that B,C,E group and A group had significant difference. Blood tests: D group and E group TC,LDL-C increased above 2 times + the other groups. Observation on ultrastructure of hippocampus CA1 area of rats under transmission electron microscope,neuronal cell ultrastructure was observed in E group the damage was most severe,followed by the B group,C group and D group again. Conclusion: The model may be used as phlegm blocking orifices of senile dementia model,used for experimental study of senile dementia.
