丹酚酸B对脑缺血再灌注损伤小鼠血管保护作用
Effects of Salvianolic Acid B on Vascular Function in Mice with Cerebral Ischemia-Reperfusion Injury
【摘要】 目的:探讨丹酚酸B对脑缺血再灌注损伤小鼠血管保护作用。方法:复制小鼠脑缺血再灌注模型,测定再灌注12 h后脑含水量和脑指数、血浆TXB2和6-keto-PGF1α含量,观察脑皮质病理学的改变及内皮P-选择素和ICAM-1的表达。结果:丹酚酸B可降低脑含水量和脑指数、血浆TXB2含量和TXB2/6-keto-PGF1α比值以及脑皮质血管内皮P-选择素和ICAM-1的表达,缓减了神经元水肿。结论:丹酚酸B对脑缺血再灌注损伤的保护作用机制,可能与其维持血管活性物质TXB2/6-keto-PGF1α平衡及降低血管内皮细胞黏附分子的表达,从而改善损伤组织局部血管功能有关。
【Abstract】 Objective:To explore the effects of Salvianolic acid B(Sal B) on vascular function in mice with cerebral ischemia-reperfusion injury.Methods:The mice model with cerebral ischemia-reperfusion was established.We investigated the water content of brain tissue,brain index and the contents of TXB2 and 6-keto-PGF1αin the plasma.The pathomorphology changes were observed and the protein expressions of P-selectin and ICAM-1 were determined.Results:Sal B decreased the content of TXB2 and TXB2/6-keto-PGF1αratio in the plasma from mice with cerebral ischemia-reperfusion injury.In addition,Sal B reduced the water content of brain tissue,brain index,the neuronal dropsy and the protein expression of P-selectin and ICAM-1 in the brain tissue.Conclusion:The mechanism that Sal B protects the cerebral ischemia-reperfusion injury may be related to maintaining the TXB2/6-keto-PGF1αhomeostasis and reducing the vascular endothelial cell adhesion molecule and improving vascular function in the ischemic tissue.
