【摘要】 目的观察不同发育时期人皮肤内表皮干细胞的分布、增殖分化特征,并探讨Integrin-β1与p53基因在皮肤发育过程中的差异性表达,以期为皮肤创伤的愈合提供进一步的理论依据。方法分别采用组织的HE、IHC和RT-PCR方法检测胎儿,少儿及成人皮肤中表皮干细胞在皮肤发育不同阶段的分布特点,以及Integrin-β1和p53在不同时期的表达情况。结果随着皮肤从胎儿、少儿到成人逐步发育成熟,表皮逐渐增厚,功能逐渐完善;Integrin-β1主要存在于皮肤基底部,其表达数量随着年龄的增大逐渐减少;p53主要分布于皮肤基底层和中层,其数量随着年龄的增大逐渐增加。RT-PCR显示Integrin-β1与p53的基因表达与IHC表达趋势一致,且胎儿与成人相比差异具有统计学意义(P<0.05)。结论随着年龄的逐渐增大,皮肤表皮的结构和功能日趋完善,Integrin-β1和p53在皮肤中的表达趋势呈负相关,这可能与不同年龄阶段皮肤损伤的修复方式不同有关。

【Abstract】 Objective In order to observe the distribution characteristics of epidermal stem cells of human skin in different developmental stages, and the differential expression of Integrin-β1 and p53 were investigated at the same time in order to provide the further theoretical basis of skin wound healing. Methods The distribution characteristics of epidermal stem cells were detdcted by HE staining. Meanwhile, IHC and RT-PCR were used to study the expression of Integrin-β1 and p53 respectively. Results The epidermis gradually became thicker and the function were gradually improving with the mature of skin from fetal to adult. The Integrin-β1mainly existed in the basal layer of the skin, and the expression gradually diminished with advancing age. The number of p53 which mainly distributed in the basal layer and the middle layer of the skin developed gradually with age. The expression trend of Integrin-β1 and p53 obtained by the RT-PCR were consistent with IHC, and showed significant difference between fetal and adult(P<0.05). Conclusion The structure and function of skin is becoming more and more perfect with age. The negative correlation of expression of Integrin- β1 and p53 may be the main reason of the reconstructive method in different age stages of skin.
