体外培育牛黄对肝内胆汁淤积大鼠雌激素受体α的影响研究

【摘要】 [目的]研究体外培育牛黄(CBS)对17α-乙炔雌二醇(EE)诱导肝内胆汁淤积(IC)大鼠体内雌激素受体α(ERα)的作用。[方法]雄性Wistar大鼠给予5 mg/kg的EE皮下注射制备IC大鼠模型,同时采用50 mg/kg和150mg/kg的CBS灌胃给药处理。连续5d后处死动物,通过HE染色、透射电镜评价CBS对IC大鼠肝脏损伤的作用;RT-PCR、western blot和免疫组织化学技术评价肝脏ERα的表达情况。[结果]CBS减轻EE诱导IC大鼠肝组织病理损伤和细胞超微结构改变,显著下调IC大鼠肝脏ERα的mRNA和蛋白表达。[结论]CBS对于EE诱导IC大鼠的保护作用可能与其减少肝脏ERα表达有关。

【Abstract】 [Objective]To study the effect of Calculus Bovis Sativus(CBS)on estrogen receptorα(ERα)in 17α-ethynylestradiol(EE)-induced intrahepatic cholestasis(IC)rat.[Methods]IC rat model was established by subcutaneous injection of EE and during modeling CBS(50or 150mg/kg per day)was intragastrically given for 5consecutive days.The animals were killed after five days and the effect of CBS on liver damage of IC rats was evaluated by HE staining and transmission electron microscopy.The expression of ERαin liver was evaluated by western blot,real-time polymerase chain reaction and immunohistochemistry.[Results]CBS attenuated the pathological damage and ultrastructural changes and decreased the expression of mRNA and protein of ERαin EE-induced IC rat liver.[Conclusion]The protective effect of CBS on EEinduced IC rat may be related to the decreased expression of ERα.
