拟南芥离体器官再生过程中CUC1的DNA甲基化修饰状态分析
DNA methylation modifi cation status of CUC1 during in vitro organogenesis in Arabidopsis thaliana
【摘要】 在植物的生长发育过程中,DNA甲基化修饰是影响基因表达的重要调控因子。CUP-SHAPED COTYLEDON 1(CUC1)是拟南芥离体器官再生的关键基因。本研究采用实时定量PCR(q RT-PCR)和亚硫酸氢盐测序PCR(BSP)技术检测了拟南芥离体器官再生过程中CUC1的表达与其DNA甲基化修饰的动态调控关系,发现CUC1的表达与其启动子区和3′端的DNA甲基化修饰水平呈明显负相关性。本研究为深入了解CUC1参与拟南芥离体器官再生的可能调控机制提供参考。
【Abstract】 DNA methylation is an important regulator of gene expression during plant development. CUPSHAPED COTYLEDON 1(CUC1) is a key marker gene during in vitro organogenesis in Arabidopsis thaliana. In this study, the dynamic relation between CUC1 expression and DNA methylation was analyzed by quantitative real-time PCR(q RT-PCR) and bisulfi te sequencing PCR(BSP), respectively, during in vitro organogenesis. The expression of CUC1 was negatively correlated with its DNA methylation level in the promoter and 3′ region. This result provides references for further understanding of the possible regulation mechanism of CUC1 in A. thaliana in vitro.
