球囊应力脑损伤大鼠模型的建立及病理观察

【摘要】 目的建立大鼠脑出血后急性应力损伤模型,观察神经功能评分及病理变化,评价该模型准确性、重复性和一致性。方法将65只雄性SD大鼠随机分为假手术组(n=15)、25μl球囊组(n=25)、50μl球囊组(n=25),分别植入50、25、50μl空瘪球囊,后两组利用压力泵10 s内充胀球囊,以模拟脑出血迅速起病。利用生存率、Longa神经功能评分、免疫组化等方法,分析不同球囊对大鼠的损伤情况。结果成功制作大鼠脑应力损伤模型。假手术组、25和50μl球囊组的24 h生存率为100%、90%、0%。与假手术组比较,25和50μl球囊组Longa神经功能评分显著提高(P<0.01),而髓鞘碱性磷酸蛋白(MBP)阳性面积明显减少(P<0.01)。25、50μl球囊组在面积损伤率、神经纤维束损伤率、MBP阳性面积上存在统计学差异(P<0.01),在尼氏体数目上没有统计学差异(P>0.01)。结论大鼠急性应力损伤模型能较准确反映脑出血的病理生理变化,且神经纤维束的损伤程度与球囊体积相关。

【Abstract】 Objective To establish a rat model of acute stress injury after intracerebral hemorrhage, observe the neurological function and pathophysiology changes, and evaluate the accuracy, repeatability and consistency of the model. Methods A total of 65 male SD rats were randomly divided into 3 groups. The rats of sham group(n = 15), 25μl balloon group(n = 25) and 50μl balloon group(n = 25)were implanted with empty balloons of 50, 25, and 50μl respectively. In order to simulate the rapid onset of intracerebral hemorrhage,the balloons were inflated by pressure pump within 10 seconds in 25 and 50μl balloon groups. The rat injury resulting from different balloons was analyzed by survival rate, Longa neurological function score and immunohistochemical staining. Results The rat model of acute brain stress injury was established successfully. The survival rates of sham, 25 and 50μl balloon groups were 100%, 90% and 0%respectively 24 h after inflation. Compared with sham group, the Longa neurological function score was increased obviously and the myelin basic protein(MBP)-positive areawas significantly decreased in 25 and 50μl balloon groups(P < 0.01). There were significant differences in the injury rate of area, injury rate of nerve fiber bundles and MBP-positive area between 25 and 50μl balloon groups(P <0.01), but there was no significant difference in the number of Nissl bodies between the two groups(P > 0.01). Conclusions The rat model of acute stress injury after intracerebral hemorrhage can precisely simulate the pathophysiologic changes in cerebral hemorrhage.The degree of stress-induced nerve fiber bundle injury is related to the volume of balloon.
