荧光定量PCR技术和分离培养法检测小肠结肠炎耶尔森菌的比较研究
Comparative study of real-time PCR and conventional culturing in the detection of Yersinia enterocolitica
【摘要】 目的比较荧光定量PCR法和分离培养法在检测小肠结肠炎耶尔森菌中特异性和灵敏度的差异。方法从686份婴幼儿腹泻患儿的粪便标本中提取细菌基因组DNA,在进行小肠结肠炎耶尔森菌保守基因fox A和黏附侵袭位点基因ail的实时荧光定量PCR检测的同时,也按常规方法进行菌株分离培养,并对2种方法的检出结果进行比较分析。结果686份标本中,分离培养法检测结果为阳性4份,阴性682份;荧光定量PCR法检测结果为阳性5份,阴性681份。荧光定量PCR法检出的5份阳性标本中,分离培养法结果为4份阳性,1份阴性。结论采用fox A联合ail基因的荧光定量PCR法特异性强,灵敏度高,且操作简便快速,检测成本低,对于监测小肠结肠炎耶尔森菌有重要意义。
【Abstract】 Objective To compare the specifility and sensitivity performances of real-time PCR and conventional culturing in the detection of Yersinia enterocolitica. Methods 686 stool samples were collected from diarrhea infants to extract bacterium genomic DNA and detect specific genes fox A and ail of Yersinia enterocolitica with real-time PCR,and isolate Yersinia enterocolitica by conventional culturing at the same time. And the detection results of the two methods were compared and analyzed. Results Among the 686 samples,4 were positive and 682 were negative in conventional culturing,and 5 were positive and 681 were negative in real-time PCR. Among the 5 real-time PCR positive samples,4 were positive and 1 was negative by conventional culturing. Conclusion The real-time PCR of fox A and ail was proved to be sensitive and specific,easy-to-use with low cost,which can be effectively employed in the surveillance of Yersinia enterocolitica.
