不同产地甘草对HepG2.2.15细胞表达乙型肝炎表面抗原和乙型肝炎e抗原的影响

【摘要】 目的通过体外实验比较不同产地的甘草提取液对乙型肝炎病毒(HBV)的抑制作用,为甘草药材的质量评价及抗病毒新药的研发提供科学依据。方法体外培养人肝癌细胞株(Hep G2.2.15)细胞,以拉米夫定作阳性对照药物,噻唑蓝(MTT)法检测甘肃、内蒙古甘草、甘草酸、甘草苷对细胞的生长抑制作用,分别计算抑制率和半数毒性浓度(TC50);酶联免疫吸附(ELISA)法检测药物对细胞表达的乙型肝炎表面抗原(HBs Ag)和乙型肝炎e抗原(HBe Ag)的影响,分别计算抑制率和半数抑制浓度(EC50),最后计算治疗指数(TI)以进行抗HBV作用评价。结果甘肃、内蒙古甘草具有抑制Hep G2.2.15细胞生长,随着药物浓度增大,其抑制率也随之增大。甘肃甘草对HBs Ag、HBe Ag抑制作用的TI值为11.570、6.760,内蒙古甘草为8.360、5.020。结论甘肃、内蒙古甘草提取液在体外均有明显抑制HBV作用,甘肃甘草作用更强。
【Abstract】 Objective To compare the inhibitory effect of different origin radix glycyrrhizae extracted solutions on hepatitis B virus( HBV) in vitro,so as to provide scientific basis for quality evaluation and antiviral drug research and development. Methods Human liver cancer cell lines( Hep G2. 2. 15) were cultivated in vitro,and thiazole blue( MTT) method was performed to detect cell growth inhibition of Gansu,Inner Mongolia radix glycyrrhizae,glycyrrhizic acid,and licorice glycosides with Lamivudine as positive control drug,then inhibition rate and half toxicity concentration( TC50) were calculated; enzyme-linked immunosorbent( ELISA) method was applied to detect the drugs effect on cells expressing hepatitis B surface antigen( HBs Ag) and hepatitis B e antigen( HBe Ag),and the effect of inhibition rate and half inhibitory concentration( EC50) were calculated,and finally the therapeutic index( TI) was calculated to conduct anti HBV effect evaluation. Results Gansu,Inner Mongolia radix glycyrrhizae could inhibit Hep G2. 2. 15 cells growth,as the drug concentration increases,the inhibition rate also will increase.The TI of Gansu radix glycyrrhizae for HBs Ag,HBe Ag inhibition were 11. 570,6. 760,and that of Inner Mongolia’s were8. 360,5. 020. Conclusion Gansu,Inner Mongolia radix glycyrrhizae extracted fluid have obvious inhibitory effect on HBV in vitro,the former effect is stronger than the other.
