【摘要】 目的建立功能型饮料中牛磺酸的柱后衍生-高效液相色谱荧光法。方法样品稀释后直接进样,柱后OPA衍生,荧光检测器检测,外标法定量。色谱条件:流动相:乙腈-水(20∶80,V/V);柱后衍生反应温度:55℃;荧光检测器激发波长:330 nm;发射波长:450 nm。结果牛磺酸的浓度为0μg/L~80μg/L时,标准曲线线性关系良好,线性相关系数(r)为0.999 9,检出限为1μg/L,相对标准偏差(RSD)为0.2%~1.0%,加标回收率为97.2%~102.8%,精密度和准确度符合检测要求。结论本方法具有选择性好、精密度高和准确度好等优点,而且由于采用荧光检测器,大大提高了灵敏度,可采取大体积稀释,更易排除其他组分的干扰,更简便,更快捷,适用于日常的检测工作中。

【Abstract】 Objective To establish the taurine- post- column derivatization- high performance liquid chromatographic fluorescence method for the determination of taurine in functional drink. Methods The samples were injected after dilution and derivatized with OPA post- column,detected with fluorescence detector. The external standard method was used for quantification.Acetonitrile- water( 20∶80,V / V) was used as mobile phase. The temperature for derivatization reaction was at 55 ℃. The excitation wavelength was set at 330 nm and the emission wavelength was set at 450 nm. Results Good linearity was obtained when the concentration of taurine was among 0 μg / L- 80 μg / L with the linear correlation coefficient( r) as 0. 999 9. The detection limit was 1 μg / L,and the relative standard derivation( RSD) was among 0. 2%- 1. 0%,and the standard recovery rate was among 97. 2%- 102. 8%,with the precision and accuracy meeting the detection requirement. Conclusion The method has good selectivity,precision and accuracy,and due to the adoption of the fluorescence detector,greatly improving the sensitivity,which can take large volume dilution,be more easy to exclude interference from other components,and can be more simple,more efficient,and suitable for routine tests.
