普瑞巴林对SNL模型大鼠镇痛作用及对脊髓背根神经节p-JNK表达的影响

【摘要】 目的:探究普瑞巴林(pregabalin,PGB)对脊神经结扎(spinal nerve ligation,SNL)模型大鼠的镇痛效果及对脊髓背根神经节(dorsal root ganglia,DRG)磷酸化氨基末端蛋白激酶(phosphorylated c-Jun N-terminal kinase,p-JNK)表达的影响。方法:将SD大鼠随机分为5组,假手术C组、SNL组、PGB治疗P1组、P2组和P3组,于术后第11天开始,PGB 3组分别每天一次灌胃PGB 15、30、60 mg/Kg,C组和SNL组给予生理盐水,连续4 d(D11-D14)。于术后第14天,测定给药前、给药后30、60、90、120、180 min各时间点的机械痛阈和热缩足反射潜伏期的变化,测定后每组随机取10只大鼠L5 DRG,免疫荧光法测定p-JNK在DRG的表达情况。结果:与C组相比,术后第14天,SNL、P1、P2、P3组的机械痛阈值和缩足反射潜伏期明显降低(P<0.01),给药后,与SNL组相比,P1、P2、P3组的机械痛阈值和缩足反射潜伏期值逐渐增加,镇痛效果在100 min左右达到最大,并呈剂量依赖。SNL组p-JNK存在高表达现象,P2组和P3组p-JNK表达明显下调。结论:普瑞巴林可以抑制脊神经结扎引起的痛敏现象,镇痛作用在给药后90120 min达到最大,并呈剂量依赖,其机制可能是通过降低p-JNK在DRG的表达而产生镇痛作用。

【Abstract】 Objective: To investigate the effect of Pregabalin on pain and expression of p-JNK in dorsal root ganglia(DRG) in rats with neuropathic pain. Methods: Adult, male, SD rats were randomly divided into five groups: sham operation group(group C), SNL group and three pregabalin treatment groups(P1, P2, P3), Pregabalin, 15, 30, 60 mg/kg in 5 ml saline, was given by intragastric infusion once a day for 4 consecutive days starting on day 11 after operation, while rats in C group and SNL group were given equal volume of saline as control. On day 14, paw withdrawal mechanical threshold(PWMT) and paw withdrawal thermal latency(PWTL) were measured at 0, 30, 60, 90, 120 and 180 min after saline or pregabalin administration. The L5 DRG was removed to examine the expression of p-JNK by immunofluorescence. Results: Compared with C group, PWMT and PWTL were markedly decreased in SNL, P1, P2 and P3 groups on the 14 th day after operation(P < 0.01). Compared with SNL group, PWMT and PWTL were slowly increased and achieved the maximum threshold about 100 min after Pregabalin administration. Expression of p-JNK in SNL group in the ipsilateral DRG was increased significantly, while expression of p-JNK was decreased significantly in P1, P2 and P3 group compared with SNL group. Conclusions: Pregabalin can relieve the neuropathic pain induced by spinal nerve ligation and exerts its maximal analgesic effect 1.52 h after treatment in a dose-dependent manner. Inhibition of p-JNK expression in DRG may contribute to the analgesia
