负载抗原的DC及DC-CIK对小细胞肺癌杀伤作用的比较研究

【摘要】 目的探讨负载抗原的树突状细胞(Dendritic cells,DC)以及DC联合CIK细胞(cytokine induced killer cells)对小细胞肺癌细胞株NCIH-446的杀伤作用,为DC、CIK应用于小细胞肺癌的治疗提供实验数据。方法体外培养DC、CIK细胞,并将NCIH-446提取的抗原负载到DC,同时将DC与CIK联合培养,将负载抗原的DC(DC-Ag)及DC-CIK与NCI446按50∶1的比例进行杀伤活性实验,检测培养不同时间的两种细胞对NCIH-446的杀伤作用。结果 DC-Ag及DC-CIK在与NCIH-446CIK共同培养2个小时就监测到了两种细胞对NCIH-446的杀伤作用,而且随着时间的推移,杀伤作用逐渐增强,DC-Ag从2小时的57.63%提高到48小时的90.78%,而DC-CIK从62.95%提高到了91.57%,显示了两种细胞强大的杀伤作用,而且这种作用随着时间的延长效果越明显。结论负载抗原的DC及DC-CIK对小细胞肺癌细胞株NCIH-446有极强的杀伤活性,为临床小细胞肺癌的治疗提供了一种新的方法。
【Abstract】 Objective Investigate the killing effect which antigen-pulsed Dendritic cells(Dendritic cells,DC)and DC united CIK cells(cytokine induced killer cells)have on small cell lung cancer cell lines NCIH-446 and provide the experimental data for the DC,CIK used in the treatment of small cell lung cancer.Methods Culture the DC,CIK cells in vitro,load the antigen extracted from NCIH-446 to DC,co-culture DC and CIK at the same time,do the experiment of killing activity making the antigen-pulsed DC(DC-ag)and DC-CIK in a 50:1properation with NCIH-446,detective killing effect of two cells in different culture time on NCIH-446.Results The killing effect which DC-Ag and DC-CIK have on NCIH-446 can be surveyed after 2hours’ co-culture and the killing effect increases gradully over time,DC-Ag increases from 57.63%to 90.78% within 46 hours and DC-CIK increases from 62.95%to 91.57%,which shows the great killing effect of the two cells,the effect becomes more obvious over time.Conclusion Antigen-pulsed DCs and DC-CIK have strong killing activity on small cell lung cell line NCIH-446,which provides a new method for the clinical treatment of small cell lung cancer.
