PRRSV特异性肽N对PRRS免疫猪IL-12分泌的影响
The effect of PRRSV-specific peptides N on the secretion of IL-12 in peripheral blood monocytes of piglet vaccinated with PRRS
【摘要】 选取体质量相同、雌雄各半、生长环境相同的乳猪26头,通过检测筛选出猪呼吸与繁殖综合征病毒(PRRSV)、圆环病毒2型(PCV2)、猪瘟病毒(CSFV)、伪狂犬病毒(PRV)阴性仔猪15头。对这些猪正常免疫PRRSV疫苗,特定时间分离外周血淋巴细胞进行培养,用PRRSV特异性肽N进行刺激,采用ELISA方法测定不同时间点淋巴细胞分泌IL-12的水平。结果显示:PRRSV免疫确切(经ELISA抗体检测呈阳性)14d时淋巴细胞用PRRSV-N-1肽刺激前期能够抑制IL-12的分泌,PRRSV免疫确切(经ELISA抗体检测呈阳性)28d时淋巴细胞用PRRSV-N-1肽能刺激IL-12的分泌量增加。表明淋巴细胞经PRRSV-N-1肽刺激后其IL-12的分泌量会受PRRSV抗体水平的影响,在抗体水平较高时促进淋巴细胞对IL-12的分泌。猪只在PRRSV免疫确切时(经ELISA抗体检测呈阳性),淋巴细胞经PRRSV-N-2肽刺激的时间点越往后其IL-12的分泌量会降低,随着时间的推移受抑制。可以得出PRRSV特异性肽N可以调节外周血淋巴细胞对IL-12的分泌,临床上可以使用PRRSV特异性肽N来增强对PRRSV的抵抗力,但其影响机制还有待研究。
【Abstract】 This study selected 26 suckling piglets with similar weight,each half of the male and female,living in the same environment.Among them,15 which were tested for PRRSV,PCV2,CSFV,and PRV negative,were used as objects of study,inoculating vaccine against PRRSV normally we lymphocytes were separated and cultureed from peripheral blood at a specific time,stimulated with peptide N of PRRSV to determinate levels of IL-12 secreted from lymphocytes at different time points by ELISA.The result shows that,at 14 dof the exact time post-Immunization with PRRSV(ELISA antibody test was positive),lymphocytes with peptide PRRSV-N-1stimulated could inhibit the secretion of IL-12 in the early days.However,at 28 dof the exact time,the secretion of IL-12 increased,indicating that the level of antibody against PRRSV could affect the secretion of IL-12 after lymphocytes are stimulated by peptide PRRSV-N-1.If the antibody keeps in a high level,lymphocytes it is easy to secret IL-12.When piglets are immunized with PRRSV exactly(ELISA antibody test was positive),the secretion of IL-12 in lymphocytes stimulated by peptide PRRSV-N-2will reduce with the time delay.PRRSV specific peptide N can be used to enhance the resistance to PRRSV,it is still a question how the peptide N influence lymphocytes secreting IL-12.
