右美托咪定混合氯胺酮对新生大鼠离体海马细胞凋亡的影响
Influence by dexmedetomidine mixed with ketamine on isolated hippocampus cells apoptosis from infant rats
【摘要】 目的探讨右美托咪定(Dex)混合氯胺酮对新生大鼠离体海马细胞凋亡的影响。方法 SD新生大鼠(出生7 d)海马神经元体外培养6 d,采用随机数字表法分为对照组(C组),100μmol/L氯胺酮组(K组),10μg/ml Dex组(D组),10μg/ml Dex混合100μmol/L氯胺酮组(DK组),各6只。孵育24 h后采用免疫组化法检测Caspase-3蛋白的表达,双染法检测神经元凋亡。结果与C组比较,K组的海马细胞凋亡率和Caspase-3表达水平升高(P<0.05);与C组比较,D组海马细胞凋亡率和Caspase-3表达水平差异无统计学意义(P>0.05);与K组比较,DK组和D组的海马细胞凋亡率和Caspase-3表达水平降低(P<0.05)。结论 Dex能减轻氯胺酮诱导的新生大鼠离体海马细胞凋亡。
【Abstract】 Objective To investigate influence by dexmedetomidine(Dex) mixed with ketamine on isolated hippocampus cells apoptosis from infant rats. Methods Hippocampal neuron from SD infant rats(7 d after birth) were taken for 6-d culture in vitro, and they were randomly divided into control group(group C), 100 μmol/L ketamine group(group K), 10 μg/ml Dex group(group D), and 10 μg/ml Dex mixed with 100 μmol/L ketamine group(group DK), with 6 rats in each group. After 24 h of incubation, immunohistochemistry method was applied to detect expression of Caspase-3 protein, and double-staining method was used to detect neuronal apoptosis. Results Comparing with group C, group K and increased hippocampus cells apoptosis rate and Caspase-3 expression level(P<0.05), and group D had no statistically significant difference of hippocampus cells apoptosis rate and Caspase-3 expression level(P>0.05). Comparing with group K, groups KD and D had reduced hippocampus cells apoptosis rates and Caspase-3 expression levels(P<0.05). Conclusion Dex can relieve ketamine-induced isolated hippocampus cells apoptosis from infant rats.
