芹菜素对大鼠骨肉瘤细胞凋亡及相关蛋白Bax,Bcl-2,Caspase-3,Caspase-9,细胞色素C的影响

【摘要】 目的:探讨芹菜素对大鼠骨肉瘤细胞凋亡及相关蛋白B细胞淋巴瘤/白血病-2(Bcl-2),Bcl-2相关X蛋白(Bax),半胱氨酸天冬氨酸蛋白酶-3(Caspase-3),Caspase-9及细胞色素C的影响。方法:SD大鼠50只,建立大鼠骨肉瘤模型,随机分为模型组,顺氯氨铂组(10 mg·kg-1),芹菜素低、中、高剂量组(1,2,4 mg·kg-1),每组10只,ig给药,连续给药30 d。模型组ig给等体积的生理盐水。末次给药24 h后,切取骨肉瘤组织,称质量。TUNEL法原位标记凋亡的骨肉瘤细胞;免疫组化法检测骨肉瘤组织中Bax,Bcl-2的蛋白表达;蛋白质免疫印迹(Western blot)法检测骨肉瘤组织中Caspase-3,Caspase-9,细胞色素C的蛋白含量。结果:与模型组比较,芹菜素低、中、高剂量组的骨肉瘤的质量降低,细胞凋亡数量明显增加,明显上调Bax,Caspase-3,Caspase-9,细胞色素C蛋白的表达,下调Bcl-2蛋白的表达(P<0.05)。结论:芹菜素可通过上调Bax,Caspase-3,Caspase-9,细胞色素C的蛋白表达,下调Bcl-2的蛋白表达,抑制骨肉瘤细胞的增殖,诱导骨肉瘤细胞凋亡,从而对骨肉瘤产生治疗和抑制作用。

【Abstract】 Objective: To study the effect of apigenin in treatment of rats osteosarcoma apoptosis and Bcell lymphoma 2( Bcl-2),Bcl-2 assaciated X protein( Bax),Caspase-3,Caspase-9 and nuclear factor-κB( NF-κB) protein expressions. Method: Totally 50 rats were randomly divided into model group,cisplatin group( 10mg·kg- 1),apigenin groups( 1,2,4 mg·kg- 1),with 10 rats in each group. The rat osteosarcoma model was made by operation. All groups were given drups for 30 days. Oteosarcoma was collected and weighed 24 h later after the last administration. TUNEL assay was adopted for in situ labeling of apoptosis of osteosarcoma cells.Immunohistochemistry was used to detect expressions of Bax,Bcl-2 in osteosarcoma. Caspase-3,Caspase-9 and cytochrome C( Cyt-C) expressions in osteosarcoma were tested using Western blot assay. Result: Compared with model group,apigenin-treated groups showed decrease in osteosarcoma weight,significant increase in the number of apoptotic cells,obvious up-regulation in Bax,Caspase-3,Caspase-9 and Cyt-C protein,and down-regulation in the expression of Bcl-2. Conclusion: Apigenin can increase Bax,Caspase-3,Caspase-9,the expression of Cyt-C,down-regulate the expression of Bcl-2,inhibit the proliferation of osteosarcoma cells,and induce apoptosis in osteosarcoma cells,so as to show a therapeutic and inhibitive effect on osteosarcoma.
