人股动脉平滑肌细胞体外培养及钙化模型的建立

【摘要】 目的:建立人股动脉平滑肌细胞体外培养及血管钙化模型。方法:运用组织块贴壁法进行人股动脉平滑肌细胞体外原代和传代培养,倒置相差显微镜和免疫荧光染色方法对培养细胞进行鉴定,并建立血管钙化模型。结果:原代和传代培养的细胞生长良好。光镜下细胞呈长梭形"峰-谷"样生长,具有典型的平滑肌细胞特征。经传代培养至第8代,细胞生长特性未见异常改变,免疫荧光染色证实为平滑肌细胞。通过钙化培养后,VON KOSSA染色进一步验证平滑肌细胞钙化。结论:组织块贴壁法可成功进行人股动脉血管平滑肌细胞的体外培养,传代后细胞生物学特征稳定,钙化培养后可建立良好的平滑肌细胞钙化模型,为心血管疾病动脉粥样硬化的机制研究奠定了实验基础。

【Abstract】 Objective:To establish human femoral artery smooth muscle cells in vitro and set up vascular calcification model.Methods:Ttissue explants adherent method was applied for the in vitro primary culture and subculture of human femorl artery smooth muscle cells.Inverted phase-contrast microscope and immune fluorescent staining method were adopted to identify the cultured cells,and then establish the vascular calcification model.Results:Cells of both primary culture and subculture grew well.Cells presented with longs pindle-shape likepeak-valley under light microscope,which have the typical characteristics of smooth muscle cells.Cells were cultivated by subculture to the 8th generation.Any abnormal changes of cell growth characteristics have not found and they were confirmed to be smooth muscle cells by Immunofluorescence.Conclusion:Explants attached method can be used successfully in vitro primary culture and sbculture of human femoral artery smooth muscle cells.The biology characteristics of cell was stable after passages, and calcification model of smooth muscle cells was set up after calcified cultivation,which laid experimental foundation for the mechanism research of cardiovascular disease atherosclerosis.
